The Preclinical Drug Efficacy Study Of Dexibuprofen-Glucosamine Hydrochloride Compound To The Osteoarthritis

Osteoarthritis(OA)is a chronic disease which is based on the degeneration lesions of articular cartilage,and its main pathological symptoms are cartilago articularis degeneration and damage,hyperostosis.The primary lesions were generally considered in the cartilage and some tissues like the synovium,subchondral bone and joint capsule,were gradually affected.There are lots of ways for the treatment of osteoarthritis,but so far,still lack of ideal drugs.Objectives:studying the treatment for the osteoarthritis,exploring well-directed interventions or methods,and detecting the treatment effect of pure medicine or combined used medicines.Methods:setting and osteoarthritis rats models,design three different drug therapies for osteoarthritis models according to the different focus for treatment(1.select nosotropic medication use of Dexibuprofen 2.select etiotropic medication use of Glucosamine hydrochloride 3.combined use of the two drugs as a new treatment),evaluating by Gotdbrg VM after undertaking drug for 120d,measuring the circumference of knee joint and taking X-ray photos to observe.Results:in the rat osteoarthritis trials,all indices were significantly different on 30d after administration,showing a good effect of the compound;on the 60d and 120 d after administration,no significant difference of the detection indicators were observed in spite of glucosamine hydrochloride administered groups;the anatomic observations and X radiographs on 120d after administration showed that compound drug have certain effect.In the rabbit osteoarthritis trials,most of the detection indicators were significantly different on 30d and 120d after administration;the anatomic observations and X radiographs on 120d after adminiatration demonstrated that compound of a middle dosage and a high dosage had better effect in each medication administration group.Conclusions:when using compound of dexibuprofen and glucosamine hydrochloride at the same time appropriately,it turns out a "treating both manifestation and root cause of disease" effect,which is effective to improve the symptoms and to slow the worsening of disease.This research indicates that joint medication is appropriate in osteoarthritis treating,and this should be further explored.
