| Kadsura coccinea is a vine of the southern genus Schisandra in the family Schisandraaceae.It is a wild fruit resource to be developed and belongs to wild rare fruit trees.In recent years,the wild resources of Kadsura coccinea have been devastated due to indiscriminate mining.At the same time,there are a series of problems in the production of Kadsura coccinea fruit,such as poor taste,few edible parts,various types,and lack of excellent varieties.Therefore,it is necessary for us to systematically collect and comprehensively evaluate Kadsura coccinea germplasm resources.We need to screen excellent individual plants with good export feeling,high edible rate,good quality,high yield and stable genetics as breeding materials.In this way,we can lay the foundation for future breeding of elite varieties.This paper takes the Kadsura coccinea as the research object.The authors used SSR fluorescent labeling and capillary electrophoresis detection technology to study the distance and genetic diversity of genetic distances among different populations of Kadsura coccinea.In this project,the yield indexes of different individual plants and the proportion of fruit parts of the Kadsura coccinea were observed,the fruit was sensory evaluated,and the main effective components and nutritional components of each part of the fruit and the root system were detected.At the same time,the author carried out high-performance liquid chromatography(HPLC)fingerprint research on peels,seeds and roots.Finally,the author uses the membership function analysis method to comprehensively evaluate the Kadsura coccinea germplasm resources.The excellent individual plants obtained from the screening and evaluation of this subject can be used as breeding materials for propagation,and can be used by researchers to carry out related breeding research.They can then carry out new or improved varieties approval work.The obtained varieties can be promoted for planting in production in the future.Below are key research findings:(1)The author collected a total of 50 individual plants of Kadsura coccinea and 5individual plants of closely related species,and the origin came from 11 different provinces,cities and counties.In the molecular marker cluster analysis map,the 55 tested individual plants were mainly divided into 8 groups.The genetic distance between different individual plants is mainly related to the species and production area.There is abundant genetic diversity among different individual plants.It is feasible to screen out excellent individual plants from the abundant Kadsura coccinea germplasm resources.There were great differences in the yield composition index and the proportion of each part of the fruit among different individual plants of Kadsura coccinea.The coefficient of variation of each index was as high as 12.81%-101.89%,with very rich genetic variation.There are high-yield and excellent provenance selection potential in Kadsura coccinea germplasm resources.In addition,it is feasible to select excellent individual plants for the purpose of development and utilization of different parts of the fruit.That is to say,it is feasible to screen out the excellent individual plants with a larger proportion of each part of the fruit.(2)There are great differences in the sensory experience and pulp sugar content of the fruit of different individual plants of the Kadsura coccinea,and the coefficient of variation of7 of the 9 indicators is more than 10%.It is feasible to screen out the fruit to give people a better sensory experience,a higher sugar content,and an excellent individual plant that is more suitable for sale as a fruit.The main effective components of various parts and roots of Kadsura coccinea fruit have great differences between different individual plants.Except for the total seed triterpenes,the coefficients of variation of other indicators were all greater than10%,and the highest even reached 72.69%.The coefficient of variation of root total lignans was even more than three times that of total triterpenes.This shows that the Kadsura coccinea has the potential to screen out germplasm resources with high content of active ingredients.It is feasible to target the excellent individual plants of extractive type or medicinal material type.The contents of 17 kinds of amino acids in Kadsura coccinea pulp vary greatly among different individual plants.The variation coefficients of all amino acids among individual plants were greater than 20%,and the variation coefficient of Asp even reached 67.94%.This indicates that the amino acids in Kadsura coccinea pulp have very rich genetic variation.The germplasm resources of Kadsura coccinea have the potential to screen out individual plants with high amino acid content.And the 17 kinds of amino acids have a strong positive correlation,it is feasible to screen out the excellent individual plants with high content of 17 kinds of amino acids in the pulp.In conclusion,it is very necessary to screen the germplasm resources of different individual plants of Kadsura coccinea,and it is feasible to screen out excellent individual plants.(3)The results of HPLC-fingerprint analysis showed that the RSD values of the relative retention time of the common peaks of the peel,seeds and roots of different individual plants were all lower than 1%.However,the RSD values of the common peak areas were between26.59% and 116.83%,of which more than half of the common peak areas had RSD values higher than 50%.This indicated that the peel,seed and root components of Kadsura coccinea were relatively consistent among different individual plants.However,the content of these components varies greatly,and it has the potential to screen out individual plants with high component content.(4)The comprehensive evaluation results of membership function analysis showed that Nos.55,12,01,15,and 40 were excellent fruit-type individual plants,and had strong advantages in planting and selling as fruits.Nos.50,10,55,07,and 04 are excellent individual plants of pericarp extraction type,with strong pericarp extraction advantages.Nos.54,47,03,04,and 09 are excellent individual plants of seed extraction type,with strong seed extraction advantages.Nos.39,46,45,09,and 03 are excellent individual plants of root medicinal material type or extraction type,and have strong medicinal or extraction advantages of the root. |
