Objective: To explore the expression differences of peroxiredoxin 6(PRDX6)in eutopic and ectopic endometrium tissues and serum of patients with ovarian endometriosis(OEM).Methods: Collect ectopic and eutopic endometrium tissue and serum specimens of 20 OEM patients,and collect normal endometrium tissue and serum specimens of 20 control women at the same time.Immunohistochemistry,Western Blot(WB),real-time fluorescence quantitative reverse transcription polymerase chain reaction(q PCR)and enzyme-linked immunosorbent assay(ELISA)were used to detect the expression of PRDX6 in the endometrium tissue and serum of OEM group and control group,respectively.Result: 1.Comparison of general data There were no statistically significant differences between the ovarian endometriosis(OEM)group and the control group in age,specimen collection period(proliferative and secretory period),and menstrual cycle days(P>0.05).2.The expression of PRDX6 protein in endometrium tissue The results of immunohistochemical experiments showed that the eutopic and ectopic endometrium tissues of the experimental group and the normal endometrium tissues of the control group all expressed PRDX6 protein.The relative gray values of the eutopic and ectopic endometrium of the experimental group and the normal endometrium of the control group were(0.06±0.02),(0.05±0.01),(0.08±0.02).The expression of PRDX6 protein was significantly different in the eutopic and ectopic endometrium of OEM patients,and the normal endometrium of the control group(F=13.496,P<0.05).he contents of PRDX6 in the eutopic and ectopic endometrium of OEM patients was significantly lower than that of normal endometrium(P<0.05),and there was no significant difference between the eutopic and ectopic endometrium in the OEM group(P<0.05).The results of the Western Blot experiment showed that the relative gray values of the eutopic and ectopic endometrium of the experimental group and the normal endometrium of the control group were(1.3±0.2),(1.2±0.4),(1.7±0.4).The expression of PRDX6 protein in the endometrium of the three groups was significantly different(F=6.119,P<0.05),and the content of PRDX6 in the eutopic and ectopic endometrium was significantly lower than that in the normal endometrium(P<0.05).3.PRDX6 m RNA expression in endometrium tissue The content of PRDX6 m RNA was significantly different in the eutopic endometrium,ectopic endometrium and normal endometrium(F=7.420,P<0.05).The PRDX6 m RNA content of eutopic endometrium and ectopic endometrium were significantly lower than normal endometrium(P<0.05),but the difference between eutopic and ectopic endometrium was not statistically significant(P>0.05).4.The expression of PRDX6 in serum There were significant differences in serum PRDX6 content between the OEM group and the control group,and the serum PRDX6 content in the OEM group was significantly higher than that in the control group(P<0.05).Conclusion: The expression of PRDX6 in the eutopic and ectopic endometrium tissues of patients with endometriosis is significantly decreased,and the serum level in the proliferative phase is increased,suggesting that the PRDX6 antioxidant factor may be involved in the occurrence and development of the disease.
