Objective:In this study, we got eutopic,ectopic endometrium of patiens with endometriosis (EMs) and normal endometrium, and we made the nude mouse model of EMs by way of allotransplantation of normal endometrium. Immunohistochemical staining was used to detect the expression of microvessel density(MVD) and CD146in eutopic, ectopic, normal endometrium and lesions of nude mice.To discuss the role in pathogenesis of overexpression of CD146in ectopic and eutopic endometrium of patients with EMs.Methods:Immunohistochemical staining was used to detect the expression of MVD and CD146in30eutopic(16proliferative phase,14secretory phase),ectopic endometrium of EMs patients and30normal endometrium (13proliferative phase,17secretory phase) of patients with Leiomyoma of uterus. The nude mouse model of EMs was made by way of allotransplantation of normal endometrium. All nude mice were divided into A,B,C group randomly.3group mice were put to death on the5th,10th,30th day after allotransplantation, and the lesions were got to be observed the growth condition and pathological changes. Immunohistochemical staining was used to detect the expression of MVD and CD146in lesions of nude mice.Results:(1) Total success rate of model was100%.(2) The MVD in eutopic,ectopic endometrium was higher than those in control group(P<0.05), while there was no statistical differences between eutopic and ectopic endometrium (P>0.05).(3) The MVD in eutopic secretory phase endometirm was higher than those in eutopic proliferative phase endometirm(P<0.01),while this differents had no statistical significance in control group(P>0.05).(4) The nude mice in A group were observed via laparotomy that most of normal endometrium with red bottom adhered to tissue of nude mice. Angiogenesis was observed around lesions under microscope,MVD=6.90±0.98; The nude mice in B group were observed via laparotomy that normal endometrium merged with nude mouse tissue,showing red saccate. Under microscope, glands in lesions grew in number and proliferated, and some of them shew mamillary proliferation, and there was obvious angiogenesis in and around lesions, MVD=21.30±0.67; The nude mice in C group were observed via laparotomy that lesions were white and stale. Under microscope, glands decreased and shriveled, and the number of microvessel were decreased, MVD=16.13±1.06, The different of MVD in lesions of3group mice had statistical significance(P<0.05).(5) The expression of CD146in eutopic,ectopic endometrium was higher than those in control group(P<0.05), while there was no statistical differences between eutopic and ectopic endometrium (P>0.05).(6) The expression of CD146in eutopic secretory phase endometirm was higher than those in eutopic proliferative phase endometirm(P<0.01),while this differents had no statistical significance in control group(P>0.05).(7) The expression of CD146in lesions of A、B、C group nude mice were (9.88±1.12)%,(20.92±1.34)%,(16.18±1.27)%, which had statistical significance (P<0.05).(8) In eutopic,ectopic and normal endometrium of control group,the expression of CD146were positively correlated with MVD (rs=0.477、0.819、0.897, P<0.01).(9) In the lesions of3group,the expression of CD146were positively correlated with MVD (rs=0.929、0.711、0.903, P<0.05)。Conclusions:By way of allotransplantation, the normal endometrium of persons was transplanted into nude mouse cavity.This method,which is reliable, has high success rate and is with significantly experimental result, can achieve dynamic observation to the process of growth of EMs and be applied into relational experiment. The expression of CD146and MVD in eutopic and ectopic endometrium of EMs patients were higher than those in normaol endometrium; The expression of CD146in nude mouse lessions correspondingly changed when lesions grew and shriveled; The expression of CD146, no matter in eutopic, ectopic and normal endometrium or in lesions of3group nude mice, was positively correlated with MVD in relevant endometrium. These point that the overexpression of CD146may be involves in the occurrence and development of the EMs through promoting angiogenesis. The role of CD146in EMs pathogenesis need further study.
