Study On The Effects Of Active Ingredients From Balanophora Laxiflora On Lung Adenocarcinoma A549 Cells And Its Application

Non-small cell lung cancer(NSCLC)accounts for about 80% of lung cancer.Lung adenocarcinoma(LUAD),having a high mortality rate,accounts for about 50% of NSCLC.Many serious adverse reactions and obvious side effects were found in the existing drugs.It is still necessary to seek safer and more effective drugs for the treatment.Balanophora laxiflora is the dried whole plant of Balanophora laxiflora Hemsl.In this paper,the extraction,separation and purification of active ingredients,structural comfirmation,preparation of formula granules,draft quality standards for medicinal materials,formula granules and standards material,in vitro inhibition of A549 cells proliferation test were studied,and the results were described as follows:1.Screened the extraction technology of Balanophora laxifloraThe optimal extraction conditions were screened out: adding 8 times the amount of 95% ethanol,refluxing extraction at 90℃ for 90 min.2.The active ingredients’ separation and purificationThe ethyl acetate phase and petroleum ether phase were identified as the active sites.The best conditions for separating the active components of Balanophora laxiflora were screened out and 5 active compounds were obtained.3.Completed the structure confirmation and standard researchThe five active compounds were confirmed as balanophorin A,balanophorin B,β-amyrin,lupeol and gallic acid.4.Completed the screening of the preparation process of the formula granules of Balanophora laxiflora.The preparation of the formula granules of Balanophora laxiflora was with 18 times the amount of water,soaked for 30 minutes,extracted for 45 minutes,3 times,filter,concentrate,dry,then mix the extract powder and mixed auxiliary materials(medicinal material powder): soluble starch(1:1))in a ratio of 2:1,an appropriate amount of 10% starch slurry to prepare a soft material that can be "hand-held into a group,lightly pressed to disperse",and then passed through a sieve for extrusion granulation,After drying at 60℃,the granules were granulated.5.Established draft quality standards for medicinal material,formula granules and standard materialsOn the basis of studying on the character identification,TLC identification,moisture and total ash content inspection,alcohol extraction and content determination of 9 batches,the draft quality standard for the medicinal material of Balanophora laxiflora was built up.On the basis of studying on character description,TLC identification,moisture and particle size inspection,and content determination of 3 batches of formula granules,a draft quality standard of formula granules was set up.On the basis of studying on the preparation,properties,identification,and purity of balanophorin A and balanophorin B,draft quality standards for the standard materials were founded.6.Completed the experiment of inhibiting the proliferation of A549 cells in vitroIt was preliminarily confirmed that Shuhuashegu formula granules had a good inhibitory effect on the proliferation of A549 cells.
