Study On The Quality Standards And Anti-inflammatory Activity Of Huangqin Fourmula Granules And Huangqin Processed Product Formula Granules

Objective The quality standard of huangqin and wine huangqin based on standard decoction was studied and formulated,and the anti-inflammatory activity of formula granules and standard decoction was compared.The theoretical basis and experimental evidence for the standardization and clinical use of formula granules were provided.Methods On the basis of high performance liquid chromatography(HPLC),based on the multi index components(baicalin,wogonoside,baicalein and wogonin),the differences in formula particles and the standard decoction of huangqin and wine huangqin and the law of chemical composition were evaluated,and the content limit and fingerprint of the formula granules were established based on the standard decoction.In the RAW 264.7 macrophage inflammatory model,the cytotoxicity and anti-inflammatory activity of formula granules and standard decoction were compared with WST-1,Griess and ELISA,and the standard of anti inflammatory activity of huangqin and wine huangqin formula granules based on standard decoction was established.Results The high performance liquid chromatography(HPLC)for the determination of baicalin,wogonoside,baicalein and wogonin in huangqin has high accuracy and good reproducibility.The content of huangqin and wine huangqin formula granules should be from 5.32%~9.88% to 4.42%~8.21% based on standard decoction.The similarity between the formula particle fingerprint and the standard decoction should be more than 0.9.The formula granules of huangqin and wine huangqin had no obvious cytotoxicity on RAW264.7 macrophages.The levels of NO,inflammatory factors TNF-α and IL-1β produced by RAW264.7 cells induced by LPS(1 mg·L-1)were significantly reduced,the inhibition rate was around 40%,and there was no significant difference compared with the same dosage of standard decoction.Conclusions Based on the standard decoction of huangqin and wine huangqin and the quality standard of fingerprints,the consistency of the chemical composition of the formula particles and the standard decoction can be ensured.According to the quality standard,the preparation of huangqin and wine huangqin in laboratory can significantly inhibit the inflammation level of macrophages,and the anti inflammatory activity in vitro is equivalent to that of standard decoction.This study can provide some reference for standardization and clinical popularization of TCM formula granules.
