Pleurotus eryngii is known as"the delicious Boletus on the grassland"and has both edible and medicinal value.The fruiting body of P.eryngii is rich in nutritional value,and has many functions,such as promoting digestion,lowering blood pressure,blood fat,cholesterol,resisting virus,resisting tumor and enhancing immunity.In this study,Plackett-Burman(PB)experiment and response surface methodology(RSM)were used to optimize the extraction conditions of P.eryngii fruiting body polysaccharide(PEBP)and the molecular weight,determined monosaccharide composition and chemical structure of PEBP.In addition,the antioxidant capacity of PEBP in vitro and the repair effect of nonylphenol(NP)on liver and spleen were also analyzed.The results were shown below:(1)The optimum extraction conditions of PEBP were extraction temperature 80,extraction time 3h,solid-liquid ratio 1:50(g/m L),ethanol concentration 86.84%,and alcohol precipitation time 17.86h,For convenient operation,after rounding the above condition parameters,the highest yield of PEBP was 1.99%,the actual yield was 1.92±0.15%.(2)Gel permeation chromatography(GPC)analysis of PEBP showed that the weight-average molecular weight(Mw)was 2.22×10~5Da.High performance liquid chromatography(HPLC)analysis showed that the PEBP primarily contained four monosaccharides including fucose(Fuc),mannose(Man),galactose(Gal)and glucose(Glc)by the mass percentages of2%,1.75%,2.59%and 93.66%with a molar ratio of 0.35:0.33:0.49:17.63.The results of Fourier transform infrared spectroscopy(FT-IR)and nuclear magnetic resonance spectroscopy(NMR)showed that PEBP had typical characteristics of polysaccharides,which was linked by a pyran polysaccharide withβ-glucoside bond.(3)When the concentration of PEBP was 1000mg/L,the scavenging abilities of PEBP on superoxide anion and hydroxyl radical were 42.58±2.88%and 50.76±2.56%,respectively.And the reducing power of PEBP was 1.69±0.19%.(4)The levels of Aspartate transaminase(AST),Alanine transaminase(ALT)and Alkaline phosphatase(ALP)in serum decreased significantly.The animal experiments results showed that H-PEBP(400mg/kg)significantly increased the levels of catalase(CAT),glutathione peroxidase,(GSH-Px)and superoxide dismutase(SOD)in liver and spleen of mice,and decreased the content of malondiadehyde(MDA).The results of ELISA displayed that PEBP significantly reduced the expression of apoptosis protein bac in mice,and increased the level of bcl-2 protein to alleviate inflammation,decreased the content of COX-2and alleviated the inflammatory response caused by NP exposure.The results of immunohistochemical test revealed that PEBP could effectively reduce the expression of inflammatory factor IL-6 in liver and spleen,and alleviate the inflammatory response.Hematoxylin and Eosin(H&E)staining indicated that PBPE could improve the inflammation degree of mice and achieve the protective effect on the body.These results indicated that PEBP had antioxidant,anti-inflammatory and apoptosis alleviating effects,and could alleviate NP toxicity to a certain extent and protect liver and spleen tissue,providing theoretical basis and reference ideas for the development of natural anti-inflammatory and liver and spleen protection drugs.
