Actinostemma lobatum Maxim,an annual herbaceous plant in the genus Zygotes of Cucurbitaceae,its growth environment requirements are not harsh,strong adaptability,widely distributed in China.Its nature is bitter,cold,with heat and detoxification,diuresis detumescence,dispel dampness effect,clinical treatment of nephritis edema,ascites swelling,snake bite and malnutrition at the beginning of the disease mainly in the form of a prescription,and it is mainly used to eat or make soap with its seeds pressed for oil,it has rich medicinal and life value.In recent years,systematic research on Actinostemma lobatum Maxim in China is few,it was found in the literature that the research on Actinostemma lobatum Maxim was mainly focused on the whole plant.Therefore,on the basis of the investigate and survey of the chemical components and pharmacological activities of the Actinostemma lobatum Maxim,and UV guided separation is adopted,this topic systematically studied the active components of the water layer extract for drying whole grass,the main purpose is to extract its active components.In this study,14 compounds were isolated from the alcohol extract of Actinostemma lobatum Maxim by reversed-phase ODS column chromatography,TLC,gelatum Sephadex LH-20,analytical type HPLC and semi-preparative HPLC and other chromatographic methods.Their structures were identified by measuring their physical and chemical properties and analyzing their spectral data(IR,NMR,MS,etc.).These compounds are tubeimoside Ⅱ(1),tubeimoside Ⅲ(2),tubeimoside Ⅴ(3),dexylosyltubeimoside Ⅲ(4),actinostemmoside A(5),actinostemmoside K(6),actinostemmoside I(7),actinostemmoside C(8),actinostemmoside J(9),actinostemmoside L(10),actinoside E(11),kaempferol(12),quercetin(13),rutin(14).Among them,compound 6 and 10 are new compounds,named as actinostemmoside K and actinostemmoside L,compound 12,13 separated from the area for the first time.Compound 6 and 10 are damarane type tetracyclic triterpenoid saponins with novel structure,and it have similar structural characteristics with known compound 7 and 9 which have strong cytotoxicity.The chemical structures of compound 6 and 10 can be compared with those of compound 7 and 9,according to its structural characteristics,the pharmacological activity study was designed to provide scientific basis for its further development and research. |
