| Corydalis edulis Maxim.is a plant of Corydalis genus belongs to Papaveraceae family.The whole plant of C.edulis is used as a Chinese herb.It is an annual gray-green herbal,wildly grown in many provinces of China,usually at an altitude of 400 to 1200 meters.C.edulis is traditionally used to clear heat and remove toxicity,moisten lung for relieving cough,stop itching and induce astringency.Recently this herb was used as antidiabetic agents in the folk.In our previous study,the extract of C.edulis was found to decrease the blood glucose level of high-fat and high-glucose diet induced ApoE-/-mice,and increase insulin secretion of HIT-T15 cells in vitro.The CH2Cl2 extract of C.edulis showed significant insulin secretion activity and the mass spectrometry data showed that its main components are alkaloids.In order to clarify the effective material basis and action mechanism of C.edulis,a systematical study was conducted on the chemical constituents and the mass spectrometry cleavage fragment analysis of main types of alkaloids of C.edulis and biological activities of C.edulis.The results are included:1.Research on the main types of alkaloids spectral characteristics of Corydalis species has been reviewed.Three hundred and seventeen alkaloids have been obtained from Corydalis species.According to structural types,they can be mainly divided into berberine-,protoberberine-,phenanthidine-,protopine-,benzylisoquinoline-,phenylisoquinoline-,spirobenzylisoquinoline-,benzoquinoisoquinoline-,apophene-,and claraine-types.Their spectroscopic data were reviewed,which will helpful for the structural elucidation of alkaloids from Corydalis species.2.The chemical constituents were separated and purified via various column chromatography including silica,sephadex LH-20,ODS,and semipreparative HPLC.As a result,twenty-nine compounds were isolated from the CH2Cl2 extract of C.edulis,including(+)-edulisine A(1a),(-)-edulisine A(1b),edulisine B(2),epi-coryximine(3),(-)-edulisine C(4a),(+)-edulisine C(4b),(-)-edulisine D(5a),(+)-edulisine D(5b),(+)-edulisine E(6a),(-)-edulisine E(6b),capnoidine(7),edulisine F(8),dihydrosanguinarine(9),8-hydroxydihydrosanguinarine(10),amottianamide(11),protopine(12),oxohydrastinine(13),nicotinamide(14),N-trans-sinapoylmethoxytyramine(15),N-trans-sinapoyltyramine(16),N-trans-p-coumaroyltyramine(17),N-trans-feruloyltyramine(18),N-cis-feruloyltyramine(19),N-cis-p-coumaroyltyramine(20),6,7-methylenedioxydihydroisocoumarin(21),medioresinol(22),syringaresinol(23),loliolide(24),and(3S,5R,6S,7E)-5,6-epoxy-3-hydroxy-7-megastigmen-9-one(25).Among them,compounds 1a/1b,2,4a/4b-6a/6b,and 8 are new compounds,compound 21 is the first report of their natural occurrence.Compounds 10,11,14-18,20,and 22-25 were isolated from Corydalis species for the first time.Compounds 3,7,9,12,13,and 19 were firstly obtained from C.edulis.3.Besides the chemical study,the insulin secretion activities of some of the isolated compounds were evaluated.The results showed that compounds 1b and 4b could improve insulin secretion in HIT-T15 cells in vitro.4.The main types of alkaloids isolated from C.edulis were analyzed by mass spectrometry,and their mass spectrometry cleavage rules were elucidated.The main chemical components of C.edulis were qualitatively identified by the mass spectrometry fragmentation,and 44 compounds were identified. |
