Alopecia is a relatively common disease in dermatology,and its incidence is increasing year by year and showing a younger trend,which is plagued the lives of more and more people.Scald is the most harmful and complex physical injury,which often causes tissue damage.The drugs of alopecia and scald have a certain effect,but the therapeutic effects on alopecia and scars is limited.Therefore,the development of new and effective drugs is a research hotspot.Dendrobium huoshanense is a species of Orchidaceae and has been used as herbal medicine in our country for hundreds of years.Dendrobium huoshanense contains active ingredients such as polysaccharides,flavonoids and alkaloids.Many studies have shown that Dendrobium huoshanense has certain therapeutic effects on diabetes,inflammation,cancer,and liver damage.However,if Dendrobium huoshanense has therapeutic effects on alopecia and scald is still unknown.In this study,constructed the mouse model of alopecia,and treated with Dendrobium huoshanense for two weeks.The results showed that Dendrobium huoshanense can promote hair growth,and improve the morphology and structure of hair follicles in alopecia mice.Mechanically,found that Dendrobium huoshanense can inhibit apoptosis by promoting expression of anti-apoptotic gene B-cell lymphoma-2(Bcl-2),activate autophagy to induce hair to the anagen phase and prolong the anagen phase by regulating expression of autophagy-related genes p62 and microtubule-associated protein 1 light chain 3-beta(LC3II),and promote hair growth by inhibiting transforming growth factorβ1(TGF-?1)in alopecia mouse.In addition,used scalded mouse model and treated mice with Dendrobium huoshanense for two weeks.The results showed that Dendrobium huoshanense can promote wound healing.The results of H&E and Masson staining indicated that Dendrobium huoshanense can promote epithelial regeneration and collagen deposition in the scalded area.Furthermore,found that Dendrobium huoshanense can promote collagen regeneration by inducing expression of ?-smooth muscle actin(?-SMA),and improve wound healing by regulating inflammation and inhibiting apoptosis.In summary,this study demonstrates that Dendrobium huoshanense can promote hair growth,improve the morphology and structure of hair follicles in alopecia mice.In addition,Dendrobium huoshanense can also promote wound healing in scald mouse model.Our research provides a theoretical basis for the treatment of alopecia and scald by Dendrobium huoshanense. |
