Dendrobium huoshanense is a traditional edible Chinese medicine herb with a variety of biological activities which can enhance immune function,provide antioxidation and anti-tumor protection,and ameliorate pulmonary function etc.In this paper,we examined the effect and underlying mechanism of Dendrobium huoshanense polysaccharide(DHP)on the cigarette smoke(CS)-exposed mice with pulmonary inflammation in vivo.The main results obtained in this study were as the follows:(1)Apparent appearance and indicators of DHP on the cigarette smoke(CS)-exposed mice: The body weight and viscera index results showed that CS-exposed mice demonstrated marked decreased in body weight and food ration as well as the damage of lung,liver,spleen and kidney as compared to the control.DHP significantly improved growth on the body weight and food ration,and inhibited viscera damage,compared with CS-exposed mice.(2)Preventive molecular mechanisms of Dendrobium huoshanense polysaccharide(DHP)on the cigarette smoke(CS)-exposed mice with pulmonary inflammation in vivo: The results of lungs stained with hematoxylin and eosin(H&E)showed that DHP could significantly decrease the inflammatory cells and the mean linear intercepts(MLI)of lung,thus alleviating the alveolar damage in response to CS.The results of macrophage CD68 stained with immunohistochemistry(IHC)showed that DHP caused a remarkable decrease in the macrophage aggregation in lung trachea.The enzyme-linked immune sorbent assay(ELISA)results showed that levels of tumor necrosis factor-α(TNF-α)and interleukin(IL)-1β in both serum and pulmonary tissue were significantly decreased after treatment with DHP.When the mice were treated with 400 mg/kg·day DHP,the levels of TNF-α and IL-1β in both serum and pulmonary tissue were decreased respectively by 13.6%,35.3%,20.2% and 21.8% than those of the model group.Western blot analysis displayed that DHP could inhibit NF-κB and MAPK activation by suppressing P65,P38 and JNK phosphorylation and decreased expression of IκB.The electrophoretic mobility shift assay(EMSA)showed that DHP could inhibit NF-κB and AP-1 DNA binding activity as compared with model group.In conclusion,DHP ameliorated apparent appearance and inhibited inflammatory factors overproduction via the regulation of inflammatory signaling,leading to the inhibiton of pulmonary inflammatory.
