Objective:To observe the improvement of Danggui Buxue Decoction on the general behavior,body weight and hematological indexes of pregnant rats with iron deficiency anemia,and to verify the hypothesis that Danggui Buxue Decoction may enhance the expression of heme and oxidase-1 in bone marrow macrophages and affect the key link of macrophage mediated "iron recycling" in the treatment of iron deficiency anemia during pregnancy,so as to enrich the Qi hyperactivity and blood production of traditional Chinese medicine,The scientific connotation of pulp filling.Method:1.Preparation of gestational iron deficiency anemia model rats: 96 SPF grade female Wistar rats weighing 180-200 g were purchased and fed adaptively for one week.According to the random number table method,they were divided into control group(n =10)and low iron group(n = 86).1.5ml blood was collected from the inner canthus vein of rats to detect hematological indexes such as hemoglobin and red blood cells;The serum ferritin(SF)was detected by ELISA,and the value was recorded as the basic value.The blank group was fed with standard feed and drinking tap water;The low iron group was fed with low iron diet,drinking deionized water,supplemented by weekly orbital bleeding,1ml / time,to avoid iron contact during feeding.Four weeks later,the anemia model was identified by orbital blood sampling method: the rats with hemoglobin(HGB)< 110 g / L and serum ferritin(SF)< 20 ng / ml in the low iron group were randomly divided into model group(M group),Danggui Buxue Decoction low-dose group(DD group),Danggui Buxue decoction medium dose group(DZ group),Danggui Buxue Decoction high-dose group(DG group),ferrous succinate group(H Group)There were 10 rats in each group:Danggui Buxue Decoction low dose group + ferrous succinate(DD + H Group),Danggui Buxue Decoction medium dose group + ferrous succinate(DZ + H Group),Danggui Buxue Decoction high dose group + ferrous succinate(DG + H Group)and control group(N Group).Forty five adult male rats of the same strain,weighing about 250 g,were purchased from the same cage according to the ratio of female: male = 2:1,The vaginal suppository was seen in the vagina of the next day or the vagina of the female rats was washed with normal saline for vaginal smear.Sperm was observed under the low power microscope(counted as gestational day 0)and the gestational period was recorded.2.Drug intervention: on the 11 th day of pregnancy,rats in the control group and model group were intragastrically intervened with normal saline,and rats in the low iron group were intragastrically intervened with their respective drug doses until the 21 st day after parturition.The hematological indexes and serum ferritin values of rats in each group on the 21 st day after parturition,and the hematological indexes of rats on the 21 st day after parturition were detected.3.After 22 days postpartum,bone marrow macrophages were isolated,induced,cultured,identified and purified.The expression of HO-1 in bone marrow macrophages was detected by Western blot.Result:1.Successful preparation of iron deficiency anemia model in pregnancy: compared with the control group,the hemoglobin(HGB)and serum ferritin(SF)of the low iron group were significantly decreased(P < 0.05);the weight of the low iron group was lower than that of the control group(P < 0.05)Compared with the control group,the rats in the low iron group were dispirited,their hair was easy to fall off,their claw nails were white,irritable,weak and prone to curl up.2.After drug intervention:(1)general behavioral observation: compared with the model group,the mental state,hair,ear and claw skin color of the rats in each group were improved in varying degrees,among which the Danggui Buxue Decoction(medium and high dose)+ ferrous succinate group rats were full of spirit,mild personality,bright hair,active activities,and ruddy ears and claws;(2)body weight: compared with the model group: the body weight of rats in each group increased after administration,and the body weight of rats in Danggui Buxue Decoction(medium and high dose)+ ferrous succinate group increased significantly(P < 0.05),with statistical significance;(3)Hematology Index + serum ferritin: compared with the model group,HGB and SF of each group increased after administration,and Danggui Buxue Decoction(medium and high dose)+ferrous succinate group increased significantly(P < 0.05),the difference was statistically significant,indicating that the treatment was effective.3.Pups:(1)general behavioral observation: the pups in the control group were active in activities,positive in sucking,gentle in character,bright and fine in hair,and ruddy in ears and claws,while the pups in the model group were dispirited,sparse in hair,negative in sucking,and pale in ears and claws;(2)weight: compared with the model group,the weight of the pups in each group increased,among which Danggui Buxue Decoction(medium and high dose)+ ferrous succinate(3)hematological indexes: compared with the model group,the HGB and SF of each group were increased,and Danggui Buxue Decoction(medium and high dose)+ ferrous succinate group were significantly increased(P < 0.05),which indicated that the treatment was effective.4.The results of Western blot showed that: compared with the model group,the expression of HO-1 was increased in all groups except the low and medium dose Danggui Buxue Decoction groups,and the expression of HO-1 in bone marrow macrophages was significantly increased after the intervention of medium and high dose Danggui Buxue Decoction plus ferrous succinate,which was similar to that in the control group(P < 0.05).5.From the perspective of economic medicine,Danggui Buxue Decoction plus ferrous succinate group has the best effect in the treatment of iron deficiency anemia during pregnancy.Conclusion:Danggui Buxue Decoction has the effect of Invigorating Qi and blood,regulating human microcirculation.Combined with ferrous succinate,Danggui Buxue Decoction can effectively improve the general behavioral,weight and hematological indexes of iron deficiency anemia model rats during pregnancy In addition,the growth rate and anemia status of the offspring may be realized by increasing the expression of HO-1 in bone marrow macrophages and accelerating macrophage mediated iron cycle.
