Expression Of Plasma CircHIPK3 In Patients With Colorectal Cancer And Its Clinical Significance

Objective:To investigate the expression level of plasma circHIPK3 in patients with colorectal cancer(CRC),and to explore the diagnostic value and clinical significance of plasma circHIPK3 in CRC.Methods:1.Detection of plasma circHIPK3 expression levels in colorectal cancer patients and healthy subjects:Peripheral blood samples from 33 patients with colorectal cancer and 33 healthy subjects were collected,and plasma circHIPK3 expression levels were detected by real-time fluorescence quantitative assay(q RT-PCR).2.To evaluate the clinical significance of plasma circHIPK3 in colorectal cancer:χ~2 test was used to analyze the correlation between plasma circHIPK3expression level and clinicopathological features of colorectal cancer.3.To explore the correlation between plasma circHIPK3 and tumor markers:Record the serum levels of FERRITIN,carcino-embryonic antigen(CEA),carbohydrate antigen 199(CA199),Carbohydrate antigen 724 (CA724)in patients with colorectal cancer and healthy subjects,and analyze the correlation between plasma circHIPK3 and serum FERRITIN,CEA, CA199,CA724 expression in CRC.4.Evaluate the diagnostic value of single detection and combined detection of plasma circHIPK3 and serum tumor markers in colorectal cancer:ROC is performed based on the expression level of plasma circHIPK3 in patients with colorectal cancer and healthy subjects,and the level of serum FERRITIN,CEA,CA199,and CA724 Curve analysis,and calculate the area under the AUC curve,sensitivity,specificity and other indicators.To evaluate the diagnostic value of single detection and combined detection of plasma circHIPK3,serum FERRITIN,CEA,CA199,CA724 in CRC.Results:1.The expression level of plasma circHIPK3 in the colorectal cancer group was significantly higher than that in the healthy control group,and the difference between the two groups was statistically significant(P<0.05).2.Plasma circHIPK3 expression is not significantly correlated with the gender,age,TNM stage,depth of invasion,and lymph node metastasis of patients with colorectal cancer,but is significantly correlated with the degree of tumor differentiation(P<0.05).3.The level of serum FERRITIN in the colorectal cancer group was significantly lower than that of the healthy control group,and the levels of CEA,CA199,and CA724 were significantly higher than those of the healthy control group.The difference was statistically significant(P<0.05).The plasma circHIPK3 and serum levels of the colorectal cancer group were significantly lower The expressions of CEA and CA199 were positively correlated(P<0.05).4.When plasma circHIPK3 and serum FERRITIN,CEA,CA199,CA724are used to detect colorectal cancer alone,plasma circHIPK3 has the greatest diagnostic value.The AUC of plasma circHIPK3 was 0.937(95CI:0.882～0.993),the sensitivity was 84.9%,and the specificity was 93.3%.CA724 has the lowest diagnostic value,with an AUC of 0.613(95CI:0.472～0.754),a sensitivity of 48.4%,and a specificity of 77.4%.In the combined detection of colorectal cancer with plasma circHIPK3 and serum FERRITIN,CEA,CA199,and CA724,the combined detection of plasma circHIPK3 and serum FERRITIN,CEA,CA199,and CA724 has the highest diagnostic value,with an AUC of 0.971(95CI:0.862～0.989)and a sensitivity of 96.8%,the specificity is 84.6%.Conclusion:1.The expression level of plasma circHIPK3 in patients with colorectal cancer is significantly higher than that in healthy people.2.The expression level of plasma circHIPK3 in patients with colorectal cancer is correlated with the degree of tumor differentiation,and the lower the degree of differentiation,the higher the expression level of circHIPK3.3.The serum FERRITIN level of patients with colorectal cancer was significantly lower than that of healthy people.The levels of CEA,CA199,and CA724 were significantly higher than those of healthy people.Plasma circHIPK3 was positively correlated with the expression of serum CEA and CA199.4.Plasma circHIPK3 has the potential to become a diagnostic marker for colorectal cancer.The combined detection of plasma circHIPK3 and serum FERRITIN,CEA,CA199,CA724 has a high diagnostic efficiency.
