Study On Process Optimization Of LignumXanthoceras Sorbifolia Compound Plaster

Objective:Based on the clinical application of senden compound plaster,this experiment uses modern research methods to promote the modernization of senden compound plaster by improving the traditional dosage form,improving the quality standard and optimizing the preparation process,so as to lay the foundation for the future marketization and productization.Research methods:1.According to the Chinese Pharmacopoeia(2015 Edition),the medicinal materials of this compound were identified by shape,size,color,surface characteristics,properties,cross section,gas and taste.TLC was used to identify quercetin,hesperidin,Pomace and strychnine.(1)Identification of quercetin:Quercetin standard solution was prepared according to thin layer chromatography(0502)of Chinese Pharmacopoeia(2015 Edition),and the test solution was aspirated for 5 minutes μ l.They were dripped on the same silica gel G plate side by side,respectively using toluene ethyl formate formic acid(6.5:3.5:1)as the bottom plate,dried until saturated,sprayed with 2%aluminum chloride ethanol solution,heated at high temperature,and observed under ultraviolet lamp.(2)Identification of Cassia obtusifolia:Quercetin standard solution was prepared according to thin layer chromatography(0502)of Chinese Pharmacopoeia(2015 Edition)μl.They were dripped on the same silica gel G plate side by side.Petroleum ether acetone(2:1)was used as the base plate respectively.After saturation,they were dried and fumigated with ammonia steam.The yellow color was Cassia obtusifolia and the red color was chrysophanol.(3)Identification of Brucella strychnine:Quercetin standard solution was prepared according to thin layer chromatography(0502)of Chinese Pharmacopoeia(2015 Edition)μ 1.They were dripped on the same silica gel G plate side by side,and each plate was made of cyclophenol dimethylamine(6:4).After saturation,they were dried in the air,sprayed with potassium iodide,and observed under the ultraviolet lamp.2.According to the Chinese Pharmacopoeia(2015 Edition),the best extraction conditions of quercetin,hesperidin,brucine and strychnine were explored through cross experiments.The contents of quercetin,hesperidin and strychnine were determined by HPLC in Chinese Pharmacopoeia(2015 Edition).3.According to the extraction process conditions of compound active ingredients and the standard of Chinese Pharmacopoeia(2015 Edition),the extraction process of compound active ingredients was prepared and identified.result:1.ldentification of quercetin,hesperidin and strychnine by TLC.The position and color of the spots are similar to the standard chromatogram.2.5441X+0.3366·r=0.9993。2.Content determination:the content of quercetin wasl.92μg/ml～40.2 μg/mlThe regression equation was2.5441X+0.3366·r=0.9993.The linear range of 0.05mg/ml～0.005mg/ml for the determination of Cassia obtusifolia was good.The linear range of strychnine was 0.05mg/ml～0.3mg/ml.3.According to the preparation method and the best preparation process of the test sample,the experimental verification was carried out at 60 times of magnification.Three groups(n=3)were repeated in parallel.According to the orthogonal optimization process evaluation method,three batches of samples were evaluated and scored,and the RSD value of the three groups was 1.42%,which proved that the optimization method was stable and reliable.Conclusion1.The single drug of the preparation completely conforms to the records of Chinese Pharmacopoeia(2015 Edition).2.The extraction process and quality level of medicinal ingredients basically meet the requirements of the main points.This method is advanced and simple,with strong specificity,good repeatability and high accuracy,which provides an effective basis for improving the quality standard of senden compound plaster.3.The process and quality of the paste basically meet the requirements.
