| The traditional Chinese medicines of Fufang Maidonggranules were inspected in the early stage,and the extraction process of Maimendong Decoction was optimized to determine the optimal extraction method.Optimize the particle formation process and determine the optimum preparation process.The standard decoction fingerprint was determined and the quality standard of the compound pellet was studied.At the same time,the prepared Maimengdong compoundgranules were compared with the standard fingerprints to study the difference,which provided a reference for the development of Maimendong compoundgranules.According to the relevant provisions of the 2015 edition of the Chinese Pharmacopoeia,the five medicinal materials Maidong,Pinellia,ginseng,Licorice and Jujube used in Maimendong compoundgranules were identified and tested to ensure that the original medicinal materials met the requirements of the Pharmacopoeia.According to the ancient dosage,refer to the information on the ancient and modem evolution of the drug taste,determine the modem dosage by modem conversion of the prescription dosage,and strictly according to the ancient decoction method,and explore the UPLC condition of the above-mentioned decoction liquid,and carry out the Maimendong soup.The establishment of a standard fingerprint.The extraction amount and the ointment rate of Russaponin in the medicinalherbs of Ophiopogon japonicus were used as reference indicators.The L9(34)orthogonal test method was used to increase the amount of water,the number of extractions and the extraction time as the influencing factors.The extraction process was optimized to determine its optimal extraction route,and the purification,concentration and drying methods and conditions were explored.The particle formability,solubility,and soft material viscosity were used as indicators to study,using the L9(34)orthogonal test method.According to the main auxiliary material ratio,the starch dextrin ratio and the concentration of the wetting agent ethanol are used as factors to optimize the particle preparation process.The quality standard of Maimendong compoundgranules was studied.After identification and testing,the five flavored medicinal materials such as Ophio pogon,Pinellia temata,Ginseng,Glycyrrhiza glabra,and Jujube all meet the requiremen ts of the 2015 Chinese Pharmacopoeia.After screening and optimization,the optimal w ater extraction process for the prescription was determined.Add water for 30minutes,add 10 times the amount of water,and cook twice for 1.5hours each time.The decoction was filtered through 12 layers of gauze,the initial pressure was 180 mbar,and the solution was concentrated under reduced pressure at 60℃ and pre-frozen at-80℃ for 12 h.The starch dextrin is mixed 1:1 uniformly,the prepared dry paste powder and the mixed auxiliary materials are added with a wetting agent 85% ethanol to make a so ft material,granulated with a 50-mesh sieve,and dried.The quality inspection of the c ompound Ophiopogon granules was carried out,and the solubility,moldability,and moi sture were in compliance with requirements.TLC identified clear spots and good resolu tion of Ophiopogon and Pinellia ternata.After exploring the chrornatographie conditions of UPLC,it was determined that the fingerprint eonditions of the standard soup of M aimendong Decoction were ACQUITY UPLC HSS T3 column(2.1mm×100mm,1.8μm),mobile phase A acetonitrile,and 0.2% phosphoric acid aqueous solution as B,wav elength 254nm,column temperature 40℃,flow rate 0.3ml·min-1.There are 21 peaks of fingerprints of 15 batches of standard soup.The sources of some of the peaks were identified,among which the chromatographic peaks 9,11,12,13,15,15,16,19,and 20 are characteristic peaks of Ophiopogon,and the peaks 1,2,and 10 are character istic peaks of Pinellia ternata.Peak 21 is a characteristic peak of licorice,peaks 3,4,5,and 6 are characteristic of ginseng,and peaks 14 and 21 are characteristic peaks of licorice.The chromatographic peaks Nos.7 and 8 are characteristic peaks of jujube.Th ecomposition information shown in the liquid phase spectrum of the compound Maido ng granules is in good agreement with the standard spectrum of the standard decoction.The UPLC conditions of Maimendong Decoction standard decoction explored in thi s experiment are stable and feasible,and provide conditions for the subsequent research of compound Maidong granules.Moreover,the optimized screening water extraction pr ocess and granule preparation process can improve the prescription extraction rate,and the granules have good formability.The fingerprints of the granules are consistent with the fingerprints of the standard soup,which indicates that the quality of the compound Maidong granules is consistent with that of the standard decoction,which provides a basis for the development and research of the compound Maidong granules. |
