Objective: To determine the distribution of estrogen receptor-α gene in Yao postmenopausal women in Guangxi Zhuang Autonomous Region,and to understand the relationship between estrogen receptor-α gene polymorphism and osteoporosis in Yao women.To determine the effect of Camellia camellia seed extract on osteoporosis.Methods:1 Effect of estrogen receptor-αgene polymorphism in postmenopausal Yao nationality women of Guangxi on postmenopausal osteoporosis5ml peripheral venous blood was collected from 612 post-menopausal women in Guangxi Yao nationality,DNA was extracted to detect the estrogen receptor gene.Serum ALP of corresponding samples was determined by kit method.The BMD of the right calcaneal was measured by ultrasonic bone density meter.Estrogen was detected by ELISA.Methods:2 Effect of camellia seed on osteoporosis in ovariectomized ratsFemale rats aged 5 months were divided into 4 experimental groups,including sham ovariectomy group(Sham),ovariectomized model group(OVX),camellia seed alcohol extract group(Ts),estradiol benzoate group(E2).At the 4th,8th,12 th and 16 th week,After conventional anesthesia,six experimental rats in each group were randomly selected and sacrificed,left heart blood and 1 cm bone tissue from right and left caput femoris were taken.Bone tissue of right caput femoris was used to observe the morphological structure,bone tissue of left caput femoris was used to measure the calcium,phosphorus and BMD.The serum was used to measure osteoprotegerin and estrogen.Results:(1)Compared with the other four genomes in the same age group,after 60 years old,P gene significantly increased alkaline phosphatase,decreased bone mineral density,increased luteinizing hormone and follicle-stimulating hormone,and decreased estradiol and progesterone(P < 0.05).Compared with the other four genomes in the same age group,after65 years old,Xx gene decreased alkaline phosphatase,increased bone mineral density,decreased luteinizing hormone and follicle-stimulating hormone,increased estradiol and progesterone(P < 0.05).(2)The bone tissue of OVX group was significantly loose under microscope,and the bone calcium,bone phosphorus,osteoprotegerin and bone mineral density were significantly lower than the other three groups(P < 0.01).The bone calcium,phosphorus,osteoprotegerin and bone mineral density of TS group were not significantly different from those of Sham group and E2 group(P > 0.05).Compared with the other three groups,serum luteinizing hormone and follicle stimulating hormone were significantly increased in OVX group,and estradiol and progesterone were significantly decreased(P < 0.05).Compared with Sham group and E2 group,there were no significant changes in luteinizing hormone,FSH,estradiol and progesterone in TS group(P > 0.05).Conclusion:(1)The P gene of estrogen receptor alpha is a predisposing gene for postmenopausal osteoporosis,and postmenopausal women with this gene should be prophylactic.However,XX gene has a higher bone density and is not prone to postmenopausal osteoporosis.(2)Camellia seed extract has estrogen-like effect;Camellia camellia seed extract can improve serum osteocalcin,osteocalcium,phosphorus,bone density and estradiol in ovariectomized rats,and has a good effect on osteoporosis.
