| Camellia oleifera belongs to the family Theaceae, its seed recorde d in the Grand Dictionary of Chinese Medicine and the South China H erbal Drugs. Camellia oleifera seed cake, together with Camellia oleifer a seed sheel, is byproducts from pressed Camellia oleifera fruit. Camelli a oleifera seed cake, as a common traditional Chinese medicine, is reco rded in Compendium of Material Medica for the treatment of acute dys pnea, coughing and removing sputum. Camellia oleifera seed shell is m ainly for industrial use. Camellia oleifera has been cultivated for more t han2300years as a special oilseed tree in China. Yet Camellia oleifera distributed mainly over the south mountain and hill region in Xiang an d Gan and the southwest China, together with a great many of its seed has been produced, its use as an anti-oxygenic or antitumor drug was little. Camellia oleifera seed has the function of antioxidantion, antibacte rial, anti-inflammatory, cure osteo porosis and some cardio-cerebrovascul ar diseases, modern pharmacological tests also revealed that some constit uents of the Camellia oleifera seed possess remarkable antioxidation and antitumor activity.By systematic solvent extraction, we separate the70%ethanol extr act of Camellia oleifera seed into petroleum part,ethyl acetate part and alcohol precipitation part.Using UV Spectrophotometry to analysis the cl earance of DPPH from every part of Camellia oleifera seed shell;using MTT assay to selecte active part of Camellia oleifera seed cake. Based on the antioxidation and antitumor activity studies of Camelli-a oleifera seed, this experiment adopts MTT assay to selecte active con s-tituents after using MCI-GEL CHP20P chromatography, gel permeatio n chromatography and silica gel column chromatography systematically i sol-ate and purify the chemical composition,with10compounds were iso lated and identified as Kaempferol,kaempferol-3-O-β-D-glucopyranoside, kae-mpferol-3-O-dirhamn oside, amyrin-3-O-glucopyran oside, spinasterol-3-0-glucopyran oside, caffeine, palmitic acid, gallic acid,28-hydroxy fri edelin,β-sitosterol.Among these,28-hydroxy friedelin was the first time f ounded in Camellia oleifera Seed Cake.Using MTT assay on cell SGC-7901,this experiment test the chemic al constituents above and finally got the2active compounds on antitum or research,they are gallic acid and28-hydroxy friedelin, whose IC50are27.0μg·mL-1,12.1μg·mL-1.The other three compounds:kaempferol,kaemp ferol-3-O-glucopyranoside and kaempferol-3-O-dirhamn oside also have s ome effect,whose IC50are76.8μg-mL-1,101.5μg·mL-1,100.7μg·mL-1. |
