Object ive:This experimental study aims to clarify the mechanism of Taohong Siwu Decoctionin treating and improving LIRI and the regulatory role of long-chain non-coding RNA H19(lncRNAH19),that is,the effect of lncRNAH19 on TNF-α,IL-1β,IL-6 role and influence.Methods:Through the study of SD rats,the model of limb ischemia-reperfusion injury was established,and they were divided into blank group,model group,Taohong Siwu Decoction group and h19 blocker group,so as to observe the effect of different treatment methods on the morphology and pathology of gastrocnemius muscle.Changes and the influence of serum TNF-α,IL-1β,IL-6 content and other indicators,and the changes of lncRNAH19mRNA expression and TNF-α,IL-1β,IL-6 levels in gastrocnemius muscle after reperfusion,in order to obtain more accurate diagnostic result.Through this experimental study,we can further understand the mechanism of Taohong Siwu Decoction in improving limb ischemia-reperfusion injury,and at the same time reveal that lncRNAH19 participates in the regulation of TNF-α,IL-1β,and IL-6.Clinical treatment provides more complete modern medical theory support.In order to study the ischemia-reperfusion injury model of 2-month-old male SD rats,we selected 78 rats,including 6 in the blank group,24 in the model group,and 24 in the Taohong Siwu Decoction group.There are 24 rats in the dose group.Except for the blank group,the rest of the groups were operated according to the corresponding method.The Taohong Siwu Decoction group would be given Taohong Siwu Decoction by gavage,while the lncRNAH19 blocker group would be pretreated by tail vein injection.The morphology and pathological changes of the gastrocnemius muscle were studied 4 hours after the operation was completed by HE staining.In addition,RT-qPCR was used to measure the expression of gastrocnemius lncRNAH19mRNA after operation(0h,2h,4h,8h),and ELISA was used to measure and analyze the contents of TNF-α,IL-1β,and IL-6 in serum.Results:The experimental results showed that through HE staining,the muscle fiber damage and inflammatory infiltration in the Taohong Siwu Siwu Decoction group and LncRNA H19 blocker group decreased compared to the model group.Through the study,①it was found that in the same time period,compared with the blank group,the expression concentrations of inflammatory factors TNF-α,IL-1β,and IL-6 in the other three groups’ serum increased(P<0.05).However,compared with the model group,both the h19 inhibitor group and the Taohong Siwu Decoction had decreased expression concentrations of inflammatory factors in the same time period(P<0.05).Among them,the h19 inhibitor group showed a more significant decrease compared with the Taohong Siwu Decoction,and the difference had statistical significance.②In addition,it was found through the study that the serum TNF-α content of the model group,h19 inhibitor group,and Taohong Siwu Decoction group all increased significantly compared with the blank group(P<0.05).Furthermore,at different time points,the serum TNF-α content of these three groups all decreased,and the h19 inhibitor group showed a more significant decrease,while the Tao Hong Si Wu Tang group also had a lower decrease than the model group,with significant statistical value(P<0.05).③Similarly,it was found through the study that the serum IL-1β content of the model group,h19 inhibitor group,and Tao Hong Si Wu Tang group all increased significantly compared with the blank group(P<0.05).Moreover,at different time points,the serum IL-1β content of these three groups all decreased,with the h19 inhibitor group having the most significant decrease and strong statistical significance(P<0.05).④Finally,it was found through statistical analysis that the serum IL-6 concentration of both the h19 inhibitor group and Tao Hong Si Wu Tang group rats was significantly higher than that of the blank group,and the difference had significant statistical value(P<0.05).In addition,at different time points,the serum IL-6 concentration of both groups of rats decreased significantly,and the difference had significant statistical meaning(P<0.05).This indicates that both h19 inhibitors and Tao Hong Si Wu Tang can significantly alleviate skeletal muscle damage caused by LIRI injury and have a certain anti-inflammatory protective effect.Conclus ion:According to the results of experimental research,Peach-blossom Four Material Decoction can reduce the levels of inflammatory factors such as TNF-α,IL-1β,and IL-6 in LIRI by regulating the expression of lncRNAH19,thereby reducing inflammatory damage,improving the morphological structure and pathology of ischemia-reperfusion injury(IRI),and alleviating and repairing tissue damage,thus improving limb IRI.LncRNAH19 plays an important role in regulating the process of limb ischemia-reperfusion in rats.It can reduce the inflammatory response of skeletal muscle injury by regulating the levels of TNF-α,IL-1β,and IL-6,and play a role in the related pathological mechanisms.LncRNA H19 is a crucial molecule in the regulation of Peach-blossom Four Material Decoction in improving rat limb IRI,and Peach-blossom Four Material Decoction can alleviate inflammatory reactions and their related pathophysiology through lncRNA H19.
