nucleus and uterine gland nucleus,the expression in the nucleus of each embryo layer and in trophoblast nucleus,The expression of RBM10 was increased in 8.5 d and decreased in 9.5 d(P<0.05).2.Affect the general condition of mice.Compared with the control group,the weight of female mice in the experimental group increased to varying degrees.The weight of the mice in the medium concentration group increased,and the weight of the mice in the high concentration group decreased(P<0.05),and the ginger extract was low.,Medium and high concentrations did not affect the activity,hair,diet,urine and feces of Kunming female mice.3.The effect of ginger extract on the estrous cycle of mice.There is no difference in the estrous cycle between the control group and the low and medium concentration group of ginger extract.The estrous cycle of mice in the high concentration group is disordered and the estrus interval is prolonged(P<0.05).4.The effect of ginger extract on the morphology of decidua.Compared with the control group,mice in the low and medium concentration group of ginger extract have more endometrial glands,obvious glandular cavity,hypertrophy of stromal cells,and more blood vessels.The above manifestations gradually increase from low to medium concentration;the number of endometrial glands in the high concentration group decreases,the gland cavity is narrow,the endometrium becomes thinner,and the hyperemia is not obvious.5.The effect of ginger extract on mouse uterine index and embryos.There was no significant difference in uterine index between the control group and the experimental group.The embryo weight in the low-medium concentration group increased from 7.5d to 9.5d,and the embryo weight in the high concentration group decreased(P<0.05).The number of mouse embryos in the 6.5d medium concentration group increased,and the number of mouse embryos in the high concentration group decreased(P<0.05).The number of mouse embryos in the 8.5d low and medium concentration groups was significantly higher than that of the control group(P<0.05).6.Immunohistochemistry of ginger extract on the expression of PI3K/AKT and RBM10 in decidual tissues.PI3 K,AKT and P-AKT are concentrated in the cytoplasm of glandular cells,epithelial cells and stromal cells of the decidua.RBM10 and P-PI3 K are expressed in Uterine gland cell nucleus,stromal cell nucleus.The expression of p-PI3 K protein in the medium concentration group from 7.5d to 9.5d was significantly increased(P<0.05),the expression of p-AKT in the low concentration group of 6.5d to 8.5d increased in the decidual cells,and it was higher at 6.5d and 9.5d.The expression level of p-AKT in the concentration group decreased(P<0.05).In decidual tissues at 6.5d,8.5d,and9.5d,the expression of RBM10 in the low,medium and high concentration groups showed a gradual upward trend,and the expression level of RBM10 in the medium and high concentration groups It was significantly higher than the control group(P<0.05).7.Western blot of ginger extract on the expression of PI3K/AKT and RBM10 in decidual tissues.In decidual tissues at 6.5d,7.5d,8.5d,9.5d,PI3 K,AKT,and RBM10 proteins were expressed in the control and experimental groups;The expression of p-AKT protein in the decidua tissue of the ginger extract low and medium concentration group from 7.5d to 8.5d was significantly higher than that in the control group(P<0.05),and the decidua tissue of the ginger extract low concentration group at 6.5d and 9.5d The protein expression of p-PI3 K and p-AKT was significantly higher than that of the control group(P<0.05),and the protein expression levels of p-PI3 K and p-AKT in the decidua tissue of the 6.5-9.5d high concentration group were significantly lower than those of the control group(P<0.05).The expression of RBM10 protein in decidua tissue from 6.5 days to 9.5 days was consistent with the results of immunohistochemistry.Conclusion:(1)Ginger extract has an effect on the development of endometrial tissue in mice.The low and medium concentration groups in this experiment can promote decidualization of the uterus,and the low concentration effect is more obvious.(2)Ginger extract can affect the blood supply of mouse endometrial tissue,estrus cycle,decidualization of decidual tissue and embryo implantation,etc.,thereby affecting conception and embryonic development.(3)The effect of ginger extract on mouse decidualization may be related to PI3K/AKT signaling pathway,and it may be that PI3K/AKT signaling pathway and RBM10 coordinately regulate endometrial decidualization. |
