Objective: The procective mechanism of ginger powder on male reproductive system injury induced by cadmium was observed by using cadmium poisoning mechanism in mouse testicular stromal cells and mouse testes.Methods: The expression of MMPs and the expression of apoptosis-related factors in testicular stromal cells were observed by immunohistochemistry,HE staining and immunoblotting in vivo and in vitro.Effects of pro-apoptotic factors Caspase-9,RBM10,CD44,inhibition of apoptosis factor BCL-2,and factors involved in cell activity,differentiation and adhesion,migration of 17αhydroxylase,eNOS and LHR after ginger powder induced cadmium induced injury Detection,TUNEL test to detect DNA apoptosis,the last line of statistical analysis of experimental data.Results : First,the testes were induced by different concentrations of cadmium,and the best time of injury was screened by MTT colorimetric method(12h).Immunohistochemical method was used to detect the protein.The results showed that the expression of Bcl-The expression of Caspase-9 and CD44 were up-regulated and decreased(P <0.05).Secondly,the final concentration of cadmium-induced injury was 10μmol / L,the effect time was12 hours,and ginger powder was added for 12 hours.The pathological changes of testis were observed by HE staining.TUNEL(17α hydroxylase,eNOS,LHR)in the detection of DNA apoptosis and immunohistochemistry in the detection of cell proliferation and apoptosis-related factors(RBM10,Caspase-9).(P<0.05),and the concentration-dependent effect was higher than that of the control group(P <0.05).The experimental results confirm that ginger has played a role in repairing the testicular injury caused by cadmium.Conclusion: From the morphological HE staining,TUNEL method to detect DNA apoptosis and the expression of various enzymes,it was proved that the activity of NO was related to the repairing effect of ginger powder.NO plays an important role in the apoptosis or proliferation of cells induced by cadmium in the testicular ligament cells.Through the in vivo and in vitro experiments of this experiment,it is considered that ginger has a most suitable concentration in the concentration of ginger and high concentration of ginger in the process of ginger to protect testicular injury caused by cadmium.It can promote cell repair.
