The Function Of RBM10 In Lung Adenocarcinoma | Posted on:2018-02-07 | Degree:Master | Type:Thesis | Country:China | Candidate:B W Shi | Full Text:PDF | GTID:2334330536486472 | Subject:Oncology | Abstract/Summary: | PDF Full Text Request | ObjectiveRBM10(RNA binding motif 10),a member of RNA binding motif family,plays an important role in a variety of biological processes.In recently researches,truncated and missense mutations of RBM10 were detected in lung adenocarcinoma,but the function of RBM10 in lung adenocarcinoma is still unclear.In our research,we explore the functions and mechanism of RBM10 in lung adenocarcinoma.MethodsWe established the RBM10 overexpressed cell line by lentivirus plasmid.Then proliferation assay and plate clone formation assay were performed to analyze the abilities of proliferation and clone formation in RBM10 overexpressed cells.Cell viability test was performed to analyze the drug resistance of A549 cells to paclitaxel.In cell scratch test and transwell invasion experiment we analyzed the abilities of migration and invasion in RBM10 overexpressed A549 cells.Telomeric repeat amplification protocol(TRAP)and southern blot were performed to analyze the telomerase activity and the telomere length in RBM10 overexpressed A549 cells.All statistical analyses were carried out using SPSS 23.0 software,measurement data were showed as mean ± SD.Analysis of variance was used in the mean comparison in groups,chi-square test was used in the comparison of count data.ResultsRBM10 inhibits proliferation and cloning formation abilities in A549 cells.RBM10 can also promote apoptosis induced by paclitaxel.Previous study showed RBM10 can interact with telomere regulatory protein,so we analyzed the influence of RBM10 in telomerase activity and telomere length.The results showed that RBM10 does not affect the telomerase activity,but significantly inhibits the telomere length.And we also found that RBM10 can increase the level of E-cadherin and reduce the level of Vimentin,and the RBM10 may inhibit the EMT to inhibit the migration and invasion.ConclusionThe RBM10 can shorten the telomere length to inhibit proliferation in lung adenocarcinoma cells.RBM10 can also inhibit the migration and invasion.These results suggested that RBM10 may function in different mechanisms in inhibiting lung adenocarcinoma proliferation and invasion,and may be an important regulatory protein in the development of lung adenocarcinoma. | Keywords/Search Tags: | lung adenocarcinoma, RBM10, proliferation, migration, invasion, telomere | PDF Full Text Request | Related items |
