Objective:To explore the intervention effects and possible mechanisms of the water extraction of Kunlun chrysanthemum ameliorate aortic endothelial dysfunction of high fat diet in rats exposed to hypoxia.Methods:Choose forty rats,male,4-6 weeks old,and randomly divide them into 4 groups(H+HFD+KLXJ)、(H+KLXJ)、(H+HFD)、(H):Kunlun chrysanthemum、hypoxia、hypoxia and Kunlun chrysanthemum、hypoxia and high-fat diet group.The rats were put in low-pressure oxygen cabin at simulated elevations of 5000 meters.According to the experimental study,the rats of 4 groups were given normal saline solution or fat emulsions by gavage per morning and normal saline solution or the water extracts of Kunlun chrysanthemum per afternoon.Intragastric dose 10ml/kg,the dosage of Kunlun chrysanthemum correspond to the raw medicine 536mg/kg.One week later,observe the changes of blood lipids,the structure and functions of aortic endothelial,the level of aortic endothelial NOS m RNA and the endothelial secretory factor among the rats of 4 groups.Results:Intervention of the water extracts of Kunlun chrysanthemum improve aortic endothelial structure damage and dysfunction in hypoxic rats with phlegm-turbidity syndrome and increased the levels of plasma total cholesterol,low density lipoprotein,decreased the levels of plasma nitric oxide,endothelial nitric oxide synthase m RNA.The water extracts of Kunlun chrysanthemum can obviously ameliorate the injury of aortic endothelial structures and endothelium-dependent vasorelaxation the rats in hypoxia and high-fat dietand increased the levels of von willebrandfactor,NOx in blood,eNOS m RNA in aorta.Conclusion:The water extracts of Kunlun chrysanthemum can obviously ameliorate the injury of aortic endothelial structures and dysfunction the rats in hypoxia and high-fat diet.The mechanisms are related to the improvement of the rats’ oxidation pressure in hypoxia and high-fat diet,the rise of eNOS m RNA in aorta and NOx in blood.
