Neurodevelopmental abnormalities are the key factors that lead to abnormal intelligence, language, behavior and cognition.Environmental factors,as one of the main pathogenic factors of neurodevelopmental disorders, will lead to abnormal behavior of offspring.Epidemiological studies have shown that Maternal Immune Activation(MIA)is significantly related to the abnormal brain development and behavior of offspring, and it is one of the important factors leading to the offspring suffering from neurodevelopmental disorders such as autism. It is worth noting that patients with neurodevelopmental disorders(such as autism)have obvious phenotypic differences in gender-dependent behaviors, and men show more explicit behavioral problems, such as aggressive activities, excessive activities and rigid behaviors, while women may have more serious internalized symptoms, such as anxiety, depression, social problems, attention and thinking problems. Therefore,it is necessary to explore the influence of MIA on behavioral phenotype of patients with neurodevelopmental disorders(autism)of different genders, and whether there is evidence of brain development and so on.Combining the results of clinical data analysis with the establishment of laboratory mouse validation model, this paper further explored the gender behavior phenotype and brain abnormal development of offspring individuals of neurodevelopmental disorders under MIA condition.Study 1: This study collected the medical records of autistic children from "Developmental Behavioral Pediatrics" of a hospital in Changchun,Jilin Province, and divided them into three groups: pregnant boys, pregnant girls, pregnant boys and pregnant girls. The scores of ABC scale and CARS scale were compared by 2×2 mixed experimental design.Study 2: In order to further verify the results of clinical data analysis, this study established MIA laboratory animal research model based on the results of clinical data analysis of study 1.MIA was established by intraperitoneal injection of lipopolysaccharide(LPS)to simulate bacterial infection in C57BL/6 mice on the 12 th day of pregnancy.By measuring the social behavior and brain length and width of adult offspring of MIA,the gender-dependent developmental abnormality of offspring under MIA was further explored. The main conclusions of this study are as follows:(1)MIA offspring have severe symptoms related to neurodevelopmental disorders(autism),especially in boys.(2)The laboratory animal research model based on the statistical results of clinical data showed that MIA caused abnormal gender-dependent emotions, working memory, brain length-width ratio and body weight in offspring mice, and the male mice were more serious, which was consistent with the results of clinical data analysis.
