Emotional development is related to the risk factors encountered by individuals at different stages of development,and various risk factors are mainly divided into genetic and environmental aspects.Genetic and environmental adverse risk factors will cause anxiety in individuals by influencing their socio-emotional development.Among them,the pregnancy environment is the main environmental factor encountered in the early stage of individual growth and development,and the unity of the pregnancy environment and the importance of the early stage of fetal brain development make the pregnancy environment an important factor affecting the emotional development of offspring.Maternal infection with the virus during pregnancy is one of the more common environmental factors during pregnancy.Based on the relevant research on the impact of maternal infection with the virus during pregnancy on the anxiety of offspring,this article adopts the method of animal research,and conducts in-depth research from three aspects: sex difference,neural mechanism and drug intervention by establishing an animal model of maternal infection with the virus during pregnancy,the specific research is as follows:The gender differences in the impact of maternal viral infection during pregnancy on offspring anxiety are inconsistent.Study 1 is a 2 * 2 double factor experimental design.By establishing a maternal viral infection model during pregnancy,the gender differences in anxiety behavior of 84 offspring mice were studied by means of open field test of animal behaviour test.The results showed that the male offspring of the viral infection group during pregnancy had significantly more anxious behaviors than the male offspring of the control group,showing obvious anxiety,while the female offspring did not show obvious anxiety.Therefore,there are sex differences in the effect of maternal infection during pregnancy on anxious behavior in offspring,and this effect is more pronounced in male offspring.The abnormal distribution of prefrontal cortex cells is one of the manifestations of dysplasia in the offspring brain.Study 2 adopted a mixed experimental design of 2 * 2 * 10,and selected 40 newborn mice.Immunohistochemical fluorescence technology was used to stain the tissue sections of the mouse prefrontal cortex,analyze the distribution of shallow neuronal cells in the prefrontal cortex of newborn offspring,and explore whether there are abnormalities in the structural development of the prefrontal cortex during maternal pregnancy when viral infection affects anxiety emotions,and explored the gender differences among them.The results showed that the distribution of shallow neuronal cells in the prefrontal cortex of newborn male offspring infected with the virus during maternal pregnancy was abnormal,showing a trend of sinking towards the interior of the cortex,with no significant impact on female offspring.Therefore,maternal viral infection during pregnancy results in abnormal development of the prefrontal cortex in the early stages of newborn male offspring.Finally,based on the inflammatory response in the womb and offspring brain caused by maternal infection with the virus during pregnancy,this study is to explore whether the suppression of inflammation can effectively intervene in the effect of maternal infection with the virus during pregnancy on the offspring.The study suppressed the inflammatory response of mother mice by injecting Gr-1 antibody,and investigated the anxiety behavior and early development of the prefrontal cortex in 61 offspring mice through open field testing and immunofluorescence experiments.It was found that inhibiting inflammation could improve the anxiety behavior of adult offspring after maternal infection with the virus during pregnancy,and save the early prefrontal cortex development abnormalities in offspring.Therefore,this study studied the sex difference characteristics,neural mechanisms and interventions of maternal viral infection affecting the anxiety behavior of offspring,and the final conclusion is as follows:(1)There was a gender difference in the anxiety behavior of offspring affected by maternal infection with the virus during pregnancy;(2)Maternal infection with the virus during pregnancy caused abnormal development of the prefrontal cortex structure in the early stage of offspring development;(3)Inhibition of inflammation can improve anxious behavior in offspring of maternal viral infection during pregnancy and rescue their early developmental abnormalities in the prefrontal cortex.
