| Objective:To detect the IL-2 and MEF2 mRNA expression levels of Xinjiang Uygur and Han people with allergic rhinitis (allergic rhinitis,AR) patients’CD4+T of lymphocytes, and whether Uygur and Han patients has difference comparison of this two kinds of target gene, to compared between the comparing two objective gene expression level, and to explore whether the two purpose gene can causes allergic rhinitis prevalence differences. Method:(1)Use 59 cases of allergic rhinitis,Uygur 27and Han 26,and 39 cases of healthy people as the research object, Uygur 20 and Han 19. The case group and control group respectively extract 5ml venous blood at the elbow crowd as experimental samples. (2)Use Immunomagnetic beads sep-aration techniques method to extract the CD4+T lymphocytes. (3)Extract cells total RNA and reverse transcription of cDNA and then application SYBR Green I real-time fluorescence quantitative RT-PCR detection of IL-2, MEF2 mRNA gene level. Results:With housekeeping GAPDH gene correction, IL-2 mRNA expression in the allergic rhinitis Group is (10.45±2.39) and in the healthy control group is (8.67±2.27). MEF2 mRNA levels in the allergic rhinitis Group is (3.83± 2.23) and in the healthy control group is (3.57±1.08). The IL-2 expression level of mRNA in allergic rhinitis group of Uygur is (10.42±1.54) in the Han nationality allergic rhinitis group is (10.48±3.07), in Han healthy control group is (8.63±2.84) and in the Uygur healthy control group is (8.70±1.65). The level of gene MEF2 mRNA in allergic rhinitis group of Uygur (3.97±1.44) in the Han nationality,allergic rhinitis group (3.68± 2.85), in Han healthy control group is (3.23±1.33) and in the Uygur healthy control group is (3.88±0.65). IL-2 mRNA gene is in allergic rhinitis higher than healthy control group,the difference was statistically significant(P<0.05), MEF2 mRNA levels in the allergic rhinitis group has no significant differences with the control group(P>0.05). The levels of IL-2 and MEF2 mRNA gene had no significant differences in Uygur and Han patients with allergic rhinitis (P>0.05). IL-2 mRNA gene in the allergic rhinitis group of Uygur and Han higher than the healthy control group, the difference was statistically significant(P<0.05). MEF2 mRNA levels in the allergic rhinitis group of Uygur and Han has no differences with the control group(P>0.05).Conclusion:IL-2 mRNA expression level in allergic rhinitis is higher than that of healthy controls(P<0.05), allergic rhinitis with gene expression levels higher than healthy control group; MEF2 mRNA expression has no obvious difference between allergic rhinitis group and healthy controls(P>0.05). MEF2 and IL-2 mRNA expression in Uygur and Han allergic rhinitis groups showed no significant differences(P>0.05). IL-2 mRNA gene in the allergic rhinitis group of Uygur and Han higher than the healthy control group, the difference was statistically significant(P<0.05). MEF2 mRNA levels in the allergic rhinitis group of Uygur and Han has no differences with the control group(P>0.05). IL-2, MEF2 mRNA gene expression differences may not the main cause Uygur and Han nationality the prevalence difference of allergic rhinitis. |
