| Objective Nonalcoholic fatty liver disease(NAFLD)is the most common chronic liver disease in the Western world.NAFLD is a genetic-environment-metabolic-related disea,genetic factors play an important role in the pathogenesis of NAFLD.Studies have shown that Bcl-2 levels were associated with the development of NAFLD.In this study,a case-control study was conducted to detect Bcl-2 gene polymorphism in 51 patients with NAFLD who were untreated and 60 healthy controls of Yichang Central People’s Hospital to investigate the association between Bcl-2 gene polymorphism and NAFLD in Han population in Yichang,Hubei Province.Methods 51 patients in the NAFLD group and 60 patients in the control group were collected as subjects.Collect general information including gender,age,height,weight,etc.At the same time,blood samples from the case group and the control group were collected,and liver function,blood glucose(GLU)and lipids were detected,and genomic DNA of the blood sample was extracted,and alleles of rs1801018 and rs4941195 loci of Bcl-2 gene in all subjects were detected by Taqman probe method,and the differences in the distribution of genotypes and alleles between the two groups were analyzed.Finally,the obtained data was analyzed by SPSS20.0 software.Results The general characteristics of the NAFLD group and the control group were compared: the body mass index(BMI)of the NAFLD group was higher than control group,and the difference was statistically significant(P<0.05).There was no statistical difference between the two groups in terms of gender and age(P<0.05).The biochemical parameters of the two groups were compared: the aminotransferase(ALT),aspartate aminotransferase(AST),and triglyceride(TG)level in the NAFLD group were higher than control group,and the difference between the two groups was statistically significant(P<0.05).GLU,total cholesterol(TC),high-density lipoprotein cholesterol(HDL),and lowdensity lipoprotein cholesterol(LDL)were not statistically significant between the two groups(P>0.05).Two sets of allelic detection: Bcl-2 gene rs1801018 locus AA,AG,GG genotype and rs 4941195 locus AA,AC,CC genotypes in the NAFLD group and the control group were not statistically different(P>0.05),rs1801018 locus: G allele carriers did not increase the risk of NAFLD compared with A allele carriers(OR =0.600,95%CI:0.301~1.195,P>0.05).rs4941195 locus: C allele carriers did not increase the risk of NAFLD compared with A allele carriers(OR = 0.403,95% CI 为 0.200~1.908,P>0.05).Conclusion There is no significant association between Bcl-2 rs1801018 and rs4941195 locus polymorphisms and NAFLD in Han population of Yichang.BMI is a dangerous factor for NAFLD,the prevalence of nonalcoholic fatty liver disease in overweight or obese people is higher than normal people. |
