Background:Non-alcoholic fatty liver disease(NAFLD)poses a serious health hazard affecting adults and children in the general population in the world-wide,especially in the West countries.And it is projected to be the leading indication for liver transplant.Various factors are known to favor nonalcoholic fatty liver disease(NAFLD),and we have been the focus on the research of pathogenesis of NAFLD causes.With the development of biological technology and statistical data and the excavation of the new technology in recent years,we have recognized gene plays an important role in the process of NAFLD.So far,GWAS studies have revealed the relationship between LYPLAL1 gene and NAFLD,no research has been carried out on the association between the polymorphism of LYPLAL1 and NAFLD in the Chinese Han population.Objective:The aim of present study was to investigate the association between the gene LYPLAL1 rs12137855 and NAFLD,and the influence on serum lipid profiles in a Chinese Han population..Methods:In this case-control cohort study,we selected a total of 298 unrelated adult subjects,including 184 unrelated Chinese patients diagnosed with NAFLD and 114 healthy controls.The subjects were collected from the Department of Gastroenterology and the Medical Center of Qingdao Municipal Hospital.Blood samples of each subject for biochemical analyses were collected and used for genetic DNA extraction,genotyping by polymerase chain reaction analysis(PCR)and tested serum lipid profiles using biochemical methods,such as Western Blot method(Western Blot)and determination of Enzyme coupling immune adsorbent(Enzyme-Linked Immunosorbnent Assay,ELISA).Statistical analyses were performed using SPSS statistical software,version 17.0 for window.Hardy-Weinberg equilibrium between expected and observed genotype distributions was assessed using the 2 test.Genotype and alleles were estimated by counting DNA sequencing and DNA distributions between NAFLD patients and controls were analyzed by Pearson’s 2 test or Fisher’s exact test where appropriate.The Baseline characteristics are shown as mean ± SD.Results:No signifiant differences in genotype and allele frequencies of LYPLAL1 rs12137855 was found between the NAFLD group and the controls group(P>0.05).Subjects with the variant LYPLAL1 rs12137855 genotype had a higher mean weight,body mass index(BMI)and low density lipoprotein(LDL)Conclusions:Our results show for the first time that LYPLAL1 gene is not associated with a risk of NAFLD development in the Chinese Han population.The variant carriers of overall subjects have signifiantly increased weight,BMI and LDL.
