“Huang pi vitex honey”(HPVH)belongs to vitex honey which is produced in Huangpi(Wuhan city Huangpi District).HPVH is one of the China’s four famous honey(vitex honey,jujube honey,acacia honey,lychee honey).In this paper,HPVH as a raw material to study its dealcohol ability on the acute alcoholism(AAI)mice,and analysis the substantial foundation of its dealcohol ability,on this basis,the effect of HPVH on the alcohol dehydrogenase(ADH)and Aldehyde dehydrogenase(ALDH)enzyme activity and expression level was investigated,and the mechanism of HPVH was expected to be revealed.In addition,the protective effects of HPVH on acute alcoholic liver injury mice were studied.The main contents and experimental results are as follows:1.preliminary investigation : the dealcohol ability and its mechanism on AAI mice of HPVH.First,a method for determination of serum ethanol content in mice by gas chromatography(GC)was established.the serum ethanol concentration(SAC)of the AAI mice in each group was measured by GC,the results showed that the SAC in the middle-dose group HPVH was significantly lower than the model group(P < 0.05),the SAC in the high-dose HPVH group was significantly lower than the model group(P<0.01),and compared with the model group,the serum ethanol clearance rates of the low,middle and high dose HPVH groups were 16.30%,47.71% and 78.37% respectively,which proved that HPVH had a great dealcohol activity and had obvious dose-effect relationship.In addition,the activity of alcohol dehydrogenase(ADH)and acetaldehyde dehydrogenase(ALDH)in the liver of each AAI mice group showed that: the ADH and ALDH activities of middle dose and high dose HPVH group were significantly higher than those of model group(P <0.05).It is indicated that one of the mechanisms of HPVH dealcohol is to accelerate the metabolism of ethanol by enhancing the activity of the two key enzymes,ADH and ALDH,and thus reducing the ethanol content in serum.2.The substantial foundation of HPVH’s dealcohol ability investigate :In order to analyze the substantial foundation of HPVH’s dealcohol ability,the total phenols and main carbohydrates were determined,the total phenols content in HPVH was160.81 μg/g by Folin-Ciocalteu colorimetric method,the contents of fructose,glucose,sucrose and maltose in HPVH were determined by HPLC-ELSD,respectively: 38.11%,34.18%,0.63%,and 0.59%.Then,the middle-dose HPVH was chosen as subject investigated,and the experimental group was set up according to the content of fructose,glucose and phenolic compounds in the middle-dose HPVH,SAC of AAI mice in each group was determined by GC.The results showed that the groups which contains fructose could significantly reduce SAC(P < 0.05)compared with model group and there are no significant difference between those groups(P > 0.05),while,the SAC of the groups not contained fructose were no significant difference(P> 0.05)compared with model group.Therefore,the main substantial foundation of HPVH’s dealcohol ability is fructose.3.The molecular mechanism of HPVH’s dealcohol ability investigate in AAI mice:The mechanism of HPVH’s dealcohol was elucidated at the molecular level by measuring the hepatic ADH and ALDH activity、 protein and gene expression of each group in AAI mice.It can be considered that the fructose in HPVH can accelerate the metabolism of ethanol in the body then decreased the SAC via increasing the hepatic ADH and ALDH protein expression、up-regulating the hepatic ADH and ALDH m RNA expression and increasing the hepatic ADH and ALDH activity.4.Protective effects of HPVH on acute alcoholic liver injury mice:The acute alcoholic liver injury model in mice was constructed.The relative biochemical parameters of serum and liver of each group were determined,and the histopathological changes of liver tissue in each group were also observed.The result shows: the protective effect of HPVH aganist acute alcohol-induced liver injury micel by increasing the body’s antioxidant level,reduce oxidative stress(increasing the GSH content,SOD activity),alleviate the degree of lipid peroxidation(inhibited the accumulation of MDA),improving ADH、ALDH activity and accelerate the metabolism of ethanol in vivo then reduce the damage of ethanol and its metabolites to the liver.
