| Excessive alcohol intake can lead to the inhibitory state of central nervous system. When alcohol absorption rate is higher than the metabolic rate, alcohol can pass through the "blood-brain" barrier, go into the brain and cause acute alcoholic intoxication. If people drinking for long term, alcohol and its metabolites as well as metabolism disturbance it caused can lead to alcohol-induced liver injury.Natto is a kind of traditional fermented food. It has much health care functions such as thrombolysis, reducing atherogenic properties, regulating blood lipid and blood pressure. It also has functions of anti-oxidation, anticancer, antibacterial, and so on. However, few researches about its protective effect on the alcohol-induced liver injury were conducted.This research studied the preventive and therapeutic effects of natto dehydro freezing powder on acute alcoholism and alcohol-induced liver injury in mice. The effects of anti-alcoholism and protective action on the liver and its mechanism were evaluated. This study could provide experimental basis for the research and development of natto related anti-alcoholism products.Our researches are summarized as the follo wings:1. The preventive and therapeutic effects of natto dehydro freezing powder on acute alcoholism in miceIn order to study the preventive and therapeutic effects of natto dehydro freezing powder (NFP) on acute alcoholism in mice, ICR mice were used as experimental material to carry out the preventive experiment, therapeutic experiment and test the activity of alcohol dehydrogenase (ADH) in liver. It was found that NFP could extend the alcohol tolerance time and shorten the sobriety time. Moreover, the preventive experiment had a better alcohol tolerance time and sobriety time than that of the therapeutic experiment. NFP could increase the ADH activity which was more significantly enhanced when treated by high NFP dosage group than that of the low NFP dosage group. These results suggested that NFP had obvious preventive and therapeutic effects on acute alcoholism in mice. Furthermore, the preventive effect of NFP was better than the therapeutic effect.2. Protective effect of natto dehydro freezing powder against alcohol-in duced liver injury in miceTo study the protective effect of natto dehydro freezing powder against alcohol-induced liver injury in mice and its mechanism,30mice were randomly divided into control group, model group and experimental group.56%alcohol at a dose of0.15m1/20g bw was administered (ig) to mice for14d to establish the acute alcoholic hepatic injury model. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activity in serum, as well as superoxide dismutase (SOD) activity and malondialdehyde (MDA) level in liver were measured. The TNF-a mRNA expression level of the liver tissue was detected by real-time reverse transcript ion-poly me rase chain reaction (real time-PCR). The results showed that Natto dehydro freezing powder treatment significantly reduced the activities of AST and ALT (P<0.01), dominantly inhibited the decrease of SOD activity and the increase of MDA level (P<0.01) in mice suffering from liver injury. In the model group, the expression of TNF-a mRNA increased to6.99times as compared with that of control group. However, the expression of TNF-a mRNA in experimental group is only22.6%of the TNF-a mRNA expression in the model group. These results suggested that Natto dehydro freezing powder had obvious protective effect on the alcohol-induced liver injury of mice. Its mechanism might be relevant to the decrease of SOD activity, the increase of MDA level and the reduction of TNF-a expression. |
