| Objective : To study the preventive and therapeutic effect of acetaldehyde dehydrogenase 2 on radiation-induced lung injury,and to study its mechanism as much as possible.Methods:We have purchased 76 male qualified SD rats from the experimental animal company,then we randomly divided all rats into five groups: normal group(n = 12),radiation-induced lung injury animal model group(n = 12),dexamethasone treatment group(n = 12),rat Alda-1 intervention group(n = 20)and rat Daidzin intervention group(n = 20).When we start experiment,4 groups of rats has except the normal group were anesthetized with phenobarbital sodium.We used CT to localization the rats,and chose the single dose of 20 gy DT γ rays were has sent out by 6X linear accelerator,the rays has irradiate whole chest of rats.normal group rats were anesthetized but we do not irradiated.When irradiation was completed,the model group rats and the normal group rats were given the same amount of physiological salt.And the other three groups of SD rats were given the same dose of drugs once a day until experiment is end.In the whole experiment,we recorded the body weight of rats,observed the hair removal of rats in the radiation field,and the changes of respiratory frequency of rats.The day of after irradiation first week and the fourth week,each group rats were killed by decapitation,and half of the rats in each group were killed each time to acquire blood sampling.After taking out the lungs of the rats,we first carefully observed the lung tissue of the rats in each group with a pair of naked eyes.Then the lung tissues of rats in each group were fixed in formalin and then stained with HE,and then we observed the histopathological changes of the rats lung tissue in each group.Results:(1)after simulating the location of rats with CT machine,we chose the single dose of 20 Gy irradiation dose to irradiate the SD rats(whole chest),we successfully established radiation-induced lung injury SD rats animal model.All groups no death.(2)normal group rats as usual.The body weight of rats in the model group gradually to loss after irradiation,and the rats exercise and diet water intake has been decreased,radiation lung injury changes such as varying degrees of hair removal and dyspnea occurred in the radiation field.One rat in the Daidzin group died,and the lung injury was the most severe garde.Dyspnea,wheezing sound and manic mental state appeared in the 4th week of the experiment.One rats has been died in dexamethasone treatment group,the rats body weight was significantly lossed,and the lung damage was slighter than Daidzin group.The activity of rats in Alda-1 group was close to normal group,and the damage of lung were late and mild.Conclusion:(1)we successfully established radiation-induced lung injury SD rats animal model after simulating the location of rats with CT machine,in single dose of 20 Gy irradiation dose(2)Alda-1 is the activity enhancer of acetaldehyde dehydrogenase 2,which can increase the activity of acetaldehyde dehydrogenase 2 by increasing the activity of acetaldehyde dehydrogenase 2.Increase the activity of acetaldehyde dehydrogenase 2 could reduce the damage of radiation-induced lung injury,which may delay / alleviate the preventive effect of radiation-induced lung injury by enhancing the body’s activite to eliminate ROS. |
