| Objective:Bladder cancer is a common malignant tumor of the genitourinary system,and its incidence and mortality are also high in my country.Its occurrence and development are regulated by a large number of complex factors,and it is not yet fully clear.However,the current treatment of bladder cancer is mainly surgery,radiotherapy and chemotherapy,which still cannot meet the needs of patients for diagnosis and treatment.Because gene targeted therapy has a very obvious effect in tumors in recent years,in order to find a better diagnosis and treatment plan for bladder cancer,we found that the expression of ID2 in bladder cancer was abnormal based on the analysis of bladder cancer case samples in the TCGA database,and further analyzed Its effect on survival and prognosis and its effect on the proliferation of bladder cancer cells through cell and animal experiments.Methods:1.Based on the bladder cancer patient data set in the TCGA database,the expression of ID2 in bladder cancer patients was analyzed through the database GEPIA2,and the difference in the expression of ID2 in bladder cancer patients was clarified,and ID2 was analyzed in the database for different tumor stages.For the expression in progress,the impact of ID2 on the survival and prognosis of bladder cancer patients was analyzed through the UALCAN database.2.IHC analysis of 70 clinical samples was performed to verify the difference in the expression of ID2 in bladder cancer tissues and paracancerous tissues.3.By constructing ID2 overexpressing T24-ID2 cell line and empty plasmid transfected T24-control line,using CCK8 method,EdU method,clone formation experiment to detect the effect of ID2 on the cell proliferation of bladder cancer.4.The T24-ID2 cell line and the T24 cell line transfected with empty plasmid were used for group culture,and the bladder cancer nude mice subcutaneous tumorigenesis experiment was performed to verify the effect of ID2 expression in bladder cancer on tumor cell proliferation.Results:1.Through the database GEPIA2 to analyze the expression of ID2 in bladder cancer,the study found that the expression level of ID2 in bladder cancer tissues was lower than that in paracancerous tissues(p<0.05),and the expression of ID2 in bladder cancer was related to tumor staging progress Related,bladder tumors are stage Ⅱ>stage Ⅲ>stage Ⅳ.At the same time,the impact of ID2 on the survival and prognosis of bladder cancer patients is analyzed through the UALCAN database,indicating that the survival prognosis of patients with high ID2 expression is higher than that with low expression.2.The results of immunohistochemistry showed that after analyzing 70 pairs of clinical samples,the study found that the expression of ID2 in patients with bladder cancer was different,and the expression of ID2 in bladder cancer tissue was lower than that in adjacent tissues(p<0.0001).3.By constructing ID2 overexpressing T24-ID2 cell line,using CCK8 method,EdU method,and cloning formation experiment to detect the effect of ID2 expression on the proliferation of bladder cancer cells.The experiments showed that:compared with the control group,ID2 overexpressing T24 cells It showed that cell proliferation was significantly inhibited(p<0.01).4.The results of subcutaneous tumor formation experiments using T24 cell lines overexpressing ID2 in nude mice showed that compared with the empty plasmid control group,the tumor volume and weight of T24 cells overexpressing ID2 were significantly smaller than those of the control group(p<0.05).Conclusion:This study confirmed that the expression level of ID2 in bladder cancer tissues was lower than that in adjacent tissues,and the analysis showed that the expression of ID2 was related to clinical stage.The higher the expression level of ID2,the lower the clinical stage of the tumor,and the better the survival prognosis.A series of cell experiments on the construction of bladder cancer T24 cell line with ID2 overexpression by lentivirus transfection and subcutaneous tumor-forming animal experiments in nude mice confirmed that in bladder cancer,ID2 is related to the proliferation of bladder cancer cells,and the targeted regulation of ID2 overexpression can inhibit the proliferation of bladder tumor cells and inhibit tumor growth.This study confirmed the difference in the expression of ID2 between cancer and paracancerous tissues in bladder cancer,and further clarified its influence on the proliferation function of bladder cancer cells,which has important clinical significance for the diagnosis,prognosis judgment and targeted therapy of bladder cancer. |
