Objective: More and more evidence shows that circular RNA(circRNA)represents a wide variety of endogenous RNA and plays a vital role in various biological processes by regulating gene expression.However,the underlying mechanism of circRNA in bladder cancer remains elusive.In our research,we found that hsa_circ_0001495 was significantly overexpressed in bladder cancer full transcriptome sequencing results.We speculated that it may affect the occurrence and development of bladder cancer.Therefore,its expression in bladder cancer tissues and cells was detected and analyzed.Its expression significance,and at the same time further study its mechanism of effect on bladder cancer through functional experiments.Methods: We used qRT-PCR to detect the expression level of hsa_circ_0001495 in bladder cancer tissue,and combined with clinical data for clinical correlation analysis.We use Sanger sequencing analysis and exonuclease digestion experiments to identify the circular structure characteristics of hsa_circ_0001495;and use nucleoplasmic separation experiments and fluorescence in situ hybridization experiments to perform hsa_circ_0001495 subcellular localization;use bioinformatics technology to predict circ_0001495 and miR-1184 and MAPK For the relationship of signal pathways in bladder cancer,the dual luciferase reporter assay was used to verify the targeted binding sites and binding effects of hsa_circ_0001495 and miR-1184.CCK8 assay,Edu and clone formation experiments were used to observe the effect of hsa_circ_0001495 on the proliferation of bladder cancer cells.Results: hsa_circ_0001495 is produced by splicing of exon 6 and exon 7 of CCNB1 transcript,and is mainly located in the cytoplasm of bladder cancer cells,and has the characteristics of resistance to exonuclease digestion.Compared with paired normal bladder tissues,the expression of hsa_circ_0001495 and CCNB1 was up-regulated significantly in bladder cancer tissues,while the expression of miR-1184 was downregulated significantly.The high expression of hsa_circ_0001495 is helpful for the early diagnosis of bladder cancer(AUC:0.739,sensitivity:0.906,specificity:0.562),and it is related to Grade(P=0.016).hsa_circ_0001495 enhanced the proliferation activity,DNA replication and synthesis ability,and clone formation ability of 5637 cells and T24 cells.hsa_circ_0001495 targets miR-1184 through sponge action,thereby regulating the activation and expression levels of p-ERK,p-JUN and p-JNK in the MAPK signaling pathway.Conclusion: hsa_circ_0001495 can be a potential marker for the diagnosis or progression of bladder cancer.hsa_circ_0001495 can target miR-1184 and inhibit its expression to promote the growth and progression of bladder cancer through the MAPK signaling pathway. |
