Objective: To explore the mechanism of action of total flavonoids from Smilax glabra on gouty nephropathy by means of network pharmacology.Methods: All the effective components of Smilax glabra were searched by TCMSP database,and the main active components of total flavonoids of Smilax glabra were screened out by combining with the relevant literature collected by How Net,and their related targets were obtained after Uni Prot check;At the same time,the related targets of gouty nephropathy were collected through Gene Cards database,OMIM database,TTD database and Drug Bank database,and the drug targets and disease targets were selected and eliminated to be the potential therapeutic targets of total flavonoids from Smilax glabra acting on gouty nephropathy Use Cytoscape3.8.0 software to build a visual network diagram of "compound-disease target";In addition,the protein interaction network between potential targets is established by STRING database and the core PPI network is selected Then Auto Dock is used for molecular docking to further verify the reliability of the combination of core targets and active ingredients;GO enrichment analysis and KEGG pathway analysis were carried out on related targets by DAVID database,and the mechanism of action of total flavonoids from Smilax glabra in treating gouty nephropathy was analyzed.Results: 10 active components of total flavonoids from Smilax glabra were screened out,and 163 related targets were selected.There are 135 targets related to gouty nephropathy,and the constructed visual network of total flavonoids from Smilax glabra-gouty nephropathy involves 17 core targets such as PTGS1,PTGS2,RELA,etc.The main biological processes involved in GO enrichment analysis include oxidative stress reaction,regulation of vascular system development,reaction to peptides,etc.The important pathways obtained by KEGG pathway analysis are mainly interleukin-17 signaling pathway,tumor necrosis factor signaling pathway,nuclear factor-κB signaling pathway,NOD-like receptor signaling pathway,and age-RAGE signaling pathway in diabetic complications.Conclusion: The total flavonoids of Smilax glabra may be involved in regulating immune response,substance metabolism,cell growth,apoptosis and other biological processes,and achieve therapeutic effect on gout nephropathy by regulating inflammatory immunity and sugar and metabolism pathways.The completion of this mechanism focuses on multi-target and multi-channel,which provides directions for new drug research and experimental research in the future,and provides new ideas for disease treatment.
