Objective: This study aims to explore the clinical medication rules of traditional Chinese medicine in the treatment of gouty nephropathy(GN)by the Ancient and Modern Medical Cloud Platform System,to find its core traditional Chinese medicine prescription,and further explore the core traditional Chinese medicine through the method of network pharmacology.The mechanism of action of prescription treatment of GN is expected to provide certain theoretical guidance and experimental research direction for the clinical treatment of GN by traditional Chinese medicine.Methods:1.The three major Chinese databases,the China National Knowledge Infrastructure(CNKI),the China Science Periodical Database(CSPD)and the Chinese Citation Database(CCD)were comprehensively searched by computer from January1,1989 to On December 17,2021,the randomized clinical controlled trials on the treatment of GN by traditional Chinese medicine were comprehensively collected,then the collected traditional Chinese medicines were analyzed by the frequency of drugs,the meridians,and the four qi and five flavors through the Ancient and Modern Medical Cloud Platform System,Excel software.Finally,the association rules and cluster analysis are used to analyze the relationship between the drugs and the formula rules between the drugs.2.Network pharmacology was used to analyze the new formula1(namely modified Simiao San formula,Smilax glabra,Bixie,Salvia,Achyranthes,Atractylodes,Phellodendron)by the method of network pharmacology in the treatment of GN.First,the active ingredients and potential targets of Smilax glabra,Bixie,Salvia,Achyranthes,Atractylodes,and Phellodendri were screened,in the TCMSP database,then the names of drug targets were standardized through the Uni Prot database,the targets of gouty nephropathy were searched in Pharm Gkb database,Gene Cards database,Drug Bank database and OMIM database;Venn diagrams of drug and disease targets was obtained by the Venny 2.1.After that,the STRING database was used to construct the core protein PPI network,and Cytoscape3.7.2 software was used to construct compound-target network diagram.Gene Ontology(GO)function enrichment and KEGG pathway analysis were carried out with the help of R language and Bioconductor bioinformatics software package,finally molecular docking was carried out through Pubchem,PDB database,Pymol,Autodocktools and other software.Results:1.A total of 7866 literatures were retrieved,158 literatures were finally included,and a total of 166 prescriptions were obtained.The high frequency traditional Chinese medicines for the treatment of GN include Smilax glabra,Bixie,Atractylodes,Salvia,Coicis Semen,etc.There are9 categories of medicinal properties,such as flat,warm,cold,slightly warm,slightly cold,cool,severe heat,heat,and severe cold.The medicines are mainly flat medicines;the tastes of medicines are mainly sweet,bitter,and acrid;the meridians of medicines are mainly distributed in the liver,spleen and Kidney Meridian;the common high frequency drugs in association analysis are Bixie-Smilax glabra,Atractylodes-Smilax glabra,Coicis Semen-Smilax glabra,etc.and through cluster analysis,modified Simiao San formula and other 2 new prescriptions were obtained.2.A total of 89 active ingredients,247 targets related to drug components,and 337 disease targets were obtained.There were 41 common targets of GN and modified Simiao San formula.Network topology analysis of potential targets of modified Simiao San formula in the treatment of gouty nephropathy by the Cytoscape 3.7.2 software,the results showed that there were 41 nodes,272 edges,Among them,26 core targets with drgree values greater than the average were PTGS2,IL1 B,PPARG,CXCL8,MAPK3,CCL2,PPARA,HMOX1,MMP9,etc.1341 biological process(BP),20 cell composition(CC),and 126 molecular functions(MF)were obtained by GO functional enrichment analysis.KEGG pathway analysis showed that there were a total of 136 signaling pathways,and the pathways with high enrichment results were AGE-RAGE signaling pathway,IL-17 signaling pathway,TNF signaling pathway,and HIF-1 signaling pathway,etc.Conclusions: 1.Through the analysis of the prescriptions of traditional Chinese medicine for gouty nephropathy,it is found that the main medicines are expelling rheumatism and dredging joint medicine,and through the cluster analysis,the modified Simiaosan prescription and other two new prescriptions are obtained.2.Through the network pharmacology analysis of modified Simiao San formula in the treatment of GN,we find that modified Simiao San formula has the characteristics of multi-flavor,multi-ingredient,multi-target and multi-channel treatment of GN,and its main mechanism may be that its effective components are quercetin,kaempferol,luteolin,etc,acting on PTGS2,IL1 B,PPARG,CXCL8 and other targets,finally AGE-RAGE,IL-17,TNF,HIF-1 and other signaling pathways are regulated to exert therapeutic effects.
