Mechanism Of Influence Of "Medicinal And Edible" Raw Hawthorn On Inflammatory-related Factors In Rats With Impaired Glucose Regulation Based On The Theory Of "Preventive Treatment Of Disease"

Objective:To observe the effect of raw hawthorn,a kind of “homological herb of Medicinal and Edible” with the taste of sour and sweet,on the Inflammatory cytokines of rats with impaired glucose regulation,exploring the mechanism of preventing and treating impaired glucose regulation of raw hawthorn,to provide new ideas and methods for clinical prevention and treatment of impaired glucose regulation,and to provide experimental basis for the idea of preventive treatment of disease in the study.Methods: 70 SPF SD rats were randomly selected into 10 control groups and fed with normal diet,the remaining rats were fed a high-fat diet.After feeding for 3 weeks,in addition to the normal control group,the other rats fasted without water for 12 h,a one-time intraperitoneal injection of streptozotocin,dissolve in 1% sodium citrate buffer,30mg/kg was performed within 30 min.After 3 days of fasting without water all day long,FBG was measured by tail vein blood.Then,20% glucose solution was given to rats in each group at 1ml/100 g,and blood glucose was measured by tail vein blood after 2h.Rat models with impaired glucose regulation were defined as those with FBG of 6.1mmol/L~7.0mmol/L and postprandial 2h blood glucose(2hPG)≥7.8mmol/L but <11.1mmol/L.50 rats with impaired glucose regulation that were successfully modeled were randomly selected and randomly assigned as: impaired glucose regulation model group,metformin group,high dose group of raw hawthorn,medium dose group of raw hawthorn,and low dose group of raw hawthorn,10 rats in each group.Drug intervention begins three days after grouping.According to the preliminary experiments,the high,medium and low dose groups of raw hawthorn were given raw hawthorn juice by gavage at 8g/kg/d,4g/kg/d and 2g/kg/d(equivalent to 10 times of the adult dose).The metformin group was given by gavage an aqueous solution of metformin at a dose of 0.12/kg/d(10 times the adult dose).Each rat in both the model group with impaired glucose regulation and the normal control group were given distilled water2 ml by gavage.The experimental animals in the group were gavaged once a day for 3 weeks.During the period of continuous administration,the animals in each group were fed with ordinary feed,and the body weight,activity,food intake and death of the rats were observed.After 3 weeks of the experiment,7 experimental animals were randomly selected from each group,fasting without water,and blood was collected from the abdominal aorta 12 hours later,ELISA was used to detect the inflammatory factor c-reactive protein(CRP)interleukin-6(IL-6)tumor necrosis factor(TNF-α)and plasminogen activator(T-PA)inhibitor of plasminogen activator(PAI)associated with inflammatory lesions.the function of beta cells of the islet and insulin(INS)indicators related to blood glucose level observed by ELISA,to observe the function of beta cells of the islet and insulin(INS)indicators related to blood glucose level.Results: Compared with the normal control group,the content of CRP、 IL-6、TNF-α in the model group was significantly increased(P﹤0.01),T-PA content was increased(P﹤0.05),INS level was decreased(P﹤0.01),and PAI content was not significantly changed(P﹥0.05),wich have no statistical significance.Compared with the model group,the content of IL-6,TNF-α in the metformin group and high dose group of raw hawthorn decreased to varying degrees(P﹤0.05),the content of CRP、T-PA in the metformin group and high dose group of raw hawthorn decreased(P﹥0.05),and there was no difference in the content of CRP,T-PA in the medium dose group of raw hawthorn and low dose group of raw hawthorn(P 0.05).The serum concentration of INS in metformin group and high dose group of raw hawthorn was increased to different degrees(P﹤0.05).In terms of PAI,there was no statistically significant difference among the groups(P﹥0.05).In terms of IL-6、TNF-α,there was no significant difference and no statistical significance between the metformin group and the high and medium dose of raw hawthorn groups(P﹥0.05).Conclusion: There is subclinical inflammatory reaction in prediabetes,the impaired phase of sugar regulation,Raw hawthorn,especially high dose has different effects on the inflammatory factor CRP、IL-6、TNF-α and Inflammatory cytokine t-pa,the effect may be achieved by lowering blood lipid,activating blood circulation and removing blood stasis to protect vascular function.Hawthorn can improve the activity of CRP、IL-6、TNF-α to increase the level of INS,then reduces insulin resistance and improves sugar regulation,to achieve the goal of preventing impaired glucose regulation developing into diabetes.Through this experimental study,the efficacy of raw hawthorn as a medicine and food homologous with acid and sweet taste herb in the prevention and treatment of impaired sugar regulation was further clarified,providing experimental basis for the idea of preventive treatment of disease in the study.
