Objective: The study of Chaoyi Qingxin Yam Decoction on improving cognitive function of rats with vascular dementia.Method: 50 male SD rats were randomly divided into 5 groups: Qingxin Yam decoction high-dose group,Qingxin Yam decoction low-dose group,Xidezhen group,sham operation group,model group,10 rats in each group.Except for the sham operation group,all groups used "two blood vessel block + sodium nitroprusside to reduce blood pressure" to establish a VD model.Qingxin Yam decoction high-dose group was given 1.5g/kg Qingxin Yam decoction solution,Qingxin Yam decoction low-dose group was given 0.375g/kg Qingxin Yam decoction solution,Xidezhen group was given 0.6mg/kg,sham operation group and model group Give 5ml/kg of normal saline.All were given by intragastric administration for 4 weeks.In the last 5days,Morris water maze was used to detect the learning and memory ability of rats,HE staining was used to observe the pathomorphological changes of the hippocampus of VD rats,and the enzyme-linked immunosorbent assay(ELISA)was used to detect the acetylcholinesterase(Ach E),5-Serotonin(5-HT)content,use the NO test kit to detect the content of nitric oxide(NO)in rat brain tissue,and observe the expression of rat brain-derived neurotrophic factor(BDNF)by immunohistochemical staining.Results:1.The results of the Morris water maze experiment showed that compared with the sham operation group,the incubation period of the model group was significantly prolonged,and there was a significant difference between the 3rd and 4th days(P<0.01).Compared with the model group,the Qingxin Yam decoction group and the Xidezhen group had significantly shorter incubation periods.The high-dose Qingxin Yam decoction group was significantly shortened on the third day(P < 0.05),and further shortened on the fourth day(P <0.01).2.Observation of pathological sections of rat brain hippocampus by HE staining showed that the nerve cells in the hippocampus of the sham operation group were arranged neatly and the nuclei were intact;the number of nerve cells in the hippocampus of the model group was reduced,the arrangement was disordered,and some cells were shrunk and fragmented Even necrosis;Compared with the model group,the Qingxin Yam decoction high-dose group,the Qingxin Yam decoction low-dose group and the Xidezhen group had an increase in the number of nerve cells in the hippocampus and the structure was complete.3.The results of the ELISA experiment showed that compared with the sham operation group,the rat serum Ach E content increased significantly(P <0.01),and the5-HT content decreased significantly(P <0.01).Compared with the model group,the Qingxin Yam decoction high-dose group was compared with that of the sham operation group.The content of Ach E in the serum of rats in the Xidezhen group decreased significantly(P <0.01);compared with the model group,the 5-HT content of the Qingxin Yam decoction high-dose group,low-dose group,and Xidezhen group increased significantly(P <0.01).4.The experimental results showed that compared with the sham operation group,the NO content in the brain tissue of the model group was significantly increased(P<0.01);compared with the model group,the NO content in the brain tissue of the Qingxin Yam decoction low-dose group was effectively reduced(P <0.05);Compared with the model group,the NO content in the brain tissue of the Qingxin Yam decoction high-dose group and Xidezhen group was significantly reduced(P <0.01).5.Immunohistochemical staining observation showed that compared with the sham operation group,the expression of BDNF in the VD model group was significantly reduced.Compared with the VD model group,the treatment groups could promote the expression of BDNF.Conclusion: Qingxin Yam Decoction can improve the learning and memory function of VD rats,and can improve the number and morphology of nerve cells in the hippocampus of VD rats.Qingxin Yam Decoction can inhibit VD rat serum Ach E activity,increase 5-HT content,reduce rat brain tissue NO content,promote BDNF expression,and then protect the brain of VD rats and improve Learning and memory function. |
