Objective: To explore the effects of two Tongqiao Yinao recipes on the learning and memory behavior of scopolamine-induced dementia model mice,and to select the prescription with better effect to study the effect of the selected prescription on cognitive function of 2-VO model rats.Methods: 1)70 male KM mice were randomly divided into 7groups,each group of 10 mice was intragastrically administered for four weeks.20 minutes before the start of the behavioral test,except for the normal group,the mice in the other administration groups were intraperitoneally injected with scopolamine hydrobromide(2 mg/Kg)to establish an acquired memory impairment model.Morris water maze test and platform jump test are used to detect spatial and passive learning and memory ability.The contents of acetylcholine(ACh),acetylcholine transferase(Ch AT)and activity of acetylcholinesterase(ACh E)in hippocampus and cortex of mice were detected by colorimetry and ELISA.2)70 SD rats were established by bilateral common golden artery ligation(2-VO).The successful rats were randomly divided into five groups according to their body weight.Every 10 mice were divided into one group and administered by gavage for four weeks.The exploration,learning and memory abilities of rats were tested through novel object recognition experiment,Y-maze experiment and autonomous activity experiment;Plasma and whole blood viscosity were detected by rheometer,and the contents of Tch,TG,LDL-C and HDLC in serum were detected,and the effect of Tongqiao Yinao Decoction on blood lipids in 2-VO model rats was explored.The levels of ACh E、ACh、Ch AT、MDA and SOD in hippocampus and cortex were detected by colorimetric method.The morphology of rat brain tissue was observed by HE staining.Results: 1)Compared with the model group,in the water maze test,the latency time of the mice in the administration group decreased,and it decreased significantly at 4 days(P<0.05),and the number of piercings increased significantly(P<0.05 or P<0.01).In the test,the latency of stepping down was significantly prolonged(P<0.01),and the number of stepping down errors was significantly reduced(P<0.01).P<0.05 or P<0.01).2)Compared with the model group,the total distance,average speed and the number of feces of the rats in the administration group were significantly reduced in the autonomous activity test(P<0.05),and the center distance and center duration were prolonged;The latency of rats was significantly shortened(P<0.05 or P<0.01),and the rate of spontaneous alternation and the proportion of time for novel arms increased(P<0.05).Significantly increased(P<0.05).Plasma viscosity,whole blood viscosity,Tch,TG and LDL-C in each administration group were significantly decreased(P<0.05),while HDL-C in low-dose Tongqiao Yinaofang group was significantly increased(P<0.05).In the detection results of brain tissue indicators,the ACh E content in the hippocampus Tongqiao Yinaojiafang high-dose group was significantly decreased,and the ACh and Ch AT contents were significantly increased(P<0.05).The content of MDA decreased significantly(P<0.05),and the content of SOD increased with no significant difference(P>0.05).HE staining results showed that a large number of neurons in the hippocampal CA1,CA2,CA3,CA4 and DG areas of the model group were necrotic,with nuclear pyknosis and lysis,enhanced eosinophilia in the cytoplasm,and a small amount of microglial infiltration;nimodipine group,Tongqiao In the low-dose Yinaojiafang group,a small amount of neuron pyknosis and deep staining of the cells were found in CA1 and CA3 areas.In the Tongqiao Yinao plus high-dose group,only a small amount of neuron pyknosis and deep staining were found in the CA1 area.Conclusion:1)Both Tongqiao and Yinao recipes can improve the short-term acquired learning and memory impairment caused by the scopolamine mouse model to a certain extent,but the results of prescription2 can show that there is a certain degree of overexcitation,so the second part of the experiment uses the prescription one.2)Tongqiao Yi Nao Recipe can improve the learning and memory ability of vascular dementia model rats,and its mechanism may be related to the improvement of abnormal rheology,blood lipid lowering,and anti-oxidation. |
