| Objective:Investigate the effective components of "Yiceng layer" transdermal absorption solution and its mechanism of intervention on synovial fibrosis.Methods:1.HPLC-Q/TOF MS-MS meth od was used to determine the effective components of "YiCeng layer" transdermal absorption solution.2.After the establishment of KOA model,the rats were treated with "YiCeng layer",and the HE staining sections of knee synoviuni of KOA group,normal group and easy layer group were compared,the protein and gene expression of NLRP3,ASC,caspase-1,cleaved-caspase-1 compared between normal rats and KOA rats.3.The protein and gene expression levels of pro-fibrogenic factors:TGF-β,TIMP1,CollA1 in knee synovial tissue of normal group,KOA group,and YiCeng layer group were compared.4.The protein and gene expression levels of inflammasome complex:NLRP3,ASC,caspase-1,cleaved-caspase-lof synovial fibroblast in control group,LPS group and easy layer group were compared.5.The protein and gene expression levels of pro-fibrogenic factors:TGF-β,TIMP1 and CollA1 in con trol group,LPS group and YiCeng layer group were comparedResults:1.HPLC-Q/TOF MS-MS showed that the main components of "YiCeng Jayer"transdermal solution were as follows:Quinic acid,Apigenin,Acacetin,Vitexilactone,Chrysin,OroxylinA,6-Methoxy wogonin,Acanthoic acid,Tetradecanoic acid,Decursin,Notopterol,Linolic acid,Palmitic acid,Octadec-9-enoic acid,Methyl palmitate,Stearic acid.2.The protein and gene expression levels of NLRP3,ASC,caspase-1 and P10 in the synovial tissue of KOA rats were significantly up regulated than those in the normal group(P<0.05);the protein and gene expression levels of NLRP3,ASC,caspase-1 and P10 in the synovial tissue of KOA rats were significantly down regulated than those in the normal group(P<0.05),3.The protein and gene expression levels of pro-fibrogenic factors:TGF-β,TIMP1,CollA1 in the synovial tissue of KOA rats were significantly up-regulated compared with those in the normal group(P<0.05);the protein and gene expression levels of TGF-β,TIMP1,Coll Aland other fibrosis related factors were significantly down regulated after "easy layer" application(P<0.05).4.The protein and gene expression levels of NLRP3,ASC,caspase-1 and P10 in LPS group were significantly higher than those in normal group(P<0.05);the protein and gene expression levels of NLRP3,ASC,caspase-1 and P10 in LPS group were significantly lower than those in normal group(P<0.05).5.The protein and gene expression levels of pro-fibrogenic factors:TGF-β,TIMP1,Coll A land other fibrosis related factors in synovial fibroblasts of LPS group were significantly up-regulated compared with those of normal group(P<0.05);the protein and gene expression levels of TGF-β,TIMP1,CollAlwere significantly down regulated after using "easy layer"transdermal solution lyophilized powder(P<0.05)Conclusion:Activation of NLRP3/Caspase-1 inflammasome is associated with the development of KOA synovial fibrosis,which is mediated by synovial inflammation.Application of "YiCeng layer" can effectively inhibit the activation of NLRP3/Caspase-1 inflammasome.2."YiCeng layer" application can improve the knee synovial fibrosis of KOA rats by inhibiting the protein and gene expression of TGF-β,TIMP1 and Co11A1. |
