Objective:to observe the activing "Yi Qi Tiao Xue,Fu Ben Pei Yuan"Moxibustion of Zhuang medicated thread on multiple infarction dementia(MID)model of rat hippocampus amino acid neurotransmitter glutamate(Glu),aspartic acid(ASP)glycine(Gly)gamma-aminobutyric acid(GABA)content in N-methyl-D aspartate receptor subtypes NR1 and brain derived neurotrophic factor(BDNF)effect Provide some experimental basis for Chinese medicine treatment of MID and the promotion effects of a simple security price optimal treatment.Methods:Seventy male Wistar rats were randomly divided into 10 normal group and60 pre-treated embolus model group by digital random method.MID model was established by microthromboembolization method in the embolus modeling group,and the rats that did not meet the dementia standard and died were excluded.The embolus modeling group was randomly divided into model group,western medicine group,medicine line moxibustion group and medicine line non-acupoint group with 10 rats each according to random number method.The medicine line moxibustion group was treated with the line point moxibustion treatment of " " Yi Qi Tiao Xue,Fu Ben Pei Yuan ",the medicine line moxibustion group was treated with cholantine hydrochloride by gavage,the medicine line non-acupoint group was treated with acupuncture under both ribs non-acupoint treatment,the normal group and the medicine line non-acupoint group were treated with the same time and the same degree of scratching stimulation as the medicine line moxibustion group.After 4 weeks of intervention,the contents of Glu,Asp,Gly and GABA in the hippocampus of rats were detected by ELISA,and the protein expressions of NR1 and BDNF in the hippocampus were detected by Western blot.Results:1.Compared with the normal group,the content of Glu Asp in the hippocampus of the model group was significantly increased(P<0.01).Compared with the model group,the content of Glu Asp in the western medicine moxibustion group was significantly decreased(P<0.01).There was no statistical difference in the content of Glu Asp between the medicine line moxibustion group and the western medicine group(P > 0.05).There was no significant difference in the content of Glu Asp between the drug line non-acupoint group and the model group(P>0.05).2.Compared with the normal group,the contents of Gly and GABA in the model group were significantly decreased,and there was no statistical difference in the contents of Gly and GABA in the drug line non-acupoint group compared with the model group(P<0.01).Compared with model group,the contents of GLY and GABA in medicine line moxibustion group and western medicine group were significantly increased(P>0.05).There were no significant differences in GLY and GABA contents between the medicine line moxibustion and the western medicine group(P > 0.05).3.NR1 protein detection: Compared with the normal group,the expression of NR1 protein in the hippocampus of the model group was significantly up-regulated(P<0.01).Compared with the model group,the expression in the hippocampal tissue of rats in the western medicine moxibustion group was significantly decreased(P<0.01).There was no statistical difference in the content between the moxibustion group and the western medicine group(P > 0.05).There was no significant difference between the non-acupoint group and the model group(P>0.05).4.Detection of BDNF protein: Compared with the normal group,the expression of BDNF protein in the hippocampus of the model group was significantly decreased(P <0.01).Compared with the model group,the expression of the western medicine moxibustion group in the hippocampal tissue was significantly up-regulated(P <0.01).here was no statistical difference in the content between the moxibustion group and the western medicine group(P > 0.05).There was no significant difference between the non-acupoint group and the model group(P>0.05).Conclusion:"Yi Qi Tiao Xue,Fu Ben Pei Yuan" Moxibustion of Zhuang medicated thread can significantly reduce the content of excitatory amino acids Glu and Asp in MID rats’ brain,increase the content of inhibitory amino acids Gly and γ-aminobutyric acid GABA and reduce the expression of NR1 protein in hippocampus."
