| Objective:Preliminary research on the clinical agreement prescription "Huashijiangzhuo" Decoction to interfere with the formation of hyperuricemia in experimental rats,to provide a certain reference for clinical application.Methods:Totally 48 SPF male SD rats were selected and randomly divided into blank group,model group,benzbromarone group(positive control drug),and "Huashi jiangzhuo" Decoction large,medium and low dose groups,each with 8 rats.The blank group did not intervene.The model group was modeled by daily subcutaneous injection of potassium oxazinate combined with gavage of ethambutol for 2 weeks.Benzbromarone group and "Huashi jiangzhuo" Decoction large,medium,and low-dose groups were treated by intragastric administration of corresponding drugs at the same time as the experimental rats were modeled.After 2 weeks,blood was collected from the abdominal aorta of each group and kidney tissue was removed.The rats were sacrificed and the blood uric acid,blood creatinine and urea nitrogen levels were detected and the pathological changes of the rat kidney tissues were observed.Results:During the experiment,1 rat in the low and medium dose groups of "Huashi jiangzhuo"Decoction died due to gastric gavage,1 rat in the medium dose group died and 1 rat in the low dose group failed to collect blood.The small and medium dose groups failed.Each lost 2 rats.Finally,the small and medium dose groups of "Huashi jiangzhuo" Decoction 6 rats in each group,and the other 8 rats in each group entered the result analysis.The results showed:(1)The blood uric acid level of the model group was significantly higher than that of the blank group(P<0.01).(2)The blood uric acid level of rats in the large-dose and medium-dose groups of "Huashi jiangzhuo" Decoction was lower than that of the model group,and the difference was statistically significant(P<0.01).(3)There was no significant difference in blood uric acid levels between the large and medium dose groups of"Huashi jiangzhuo" Decoction and the positive control drug benzbromarone group(P>0.05).(4)Comparing the blood uric acid level of each dose group of "Huashi jiangzhuo" Decoction,there was a significant difference between the low dose,the medium dose,and the large dose(P<0.05),and the difference between the medium dose and the large dose was not significant(P>0.05).(5)Compared with the blank group,the blood creatinine and urea nitrogen levels of the model group were higher(P<0.01).Large and medium doses of "Huashi jiangzhuo" Decoction can effectively inhibit the increase of serum creatinine and urea nitrogen levels in experimental rats(P<0.05),while the effect of low dose to inhibit the increase of serum creatinine is not obvious(P>0.05).(6)The pathological results showed that "Huashi jiangzhuo" Decoction can improve the edema of renal tubular epithelial cells in experimental rats,reduce the infiltration of inflammatory cells and the proliferation of collagen fibers,and its effect is positively related to the dose.Conclusions:Both large and medium doses of "Huashi jiangzhuo" Decoction can inhibit the formation of hyperuricemia in experimental rats to a certain extent,inhibit the increase of blood creatinine and urea nitrogen levels in experimental rats,and improve the pathological changes of kidney tissue damage during the model of experimental rats in hyperuricemia formation process. |
