Effects Of Rubusoside On AMPK/GLUT2/4 Pathway And Oxidative Stress In Type 2 Diabetic Mice

Objective To investigate the hypoglycemic effect of Rub on T2 DM mice and its mechanism,and to provide theoretical basis for clinical prevention and treatment of T2 DM.Methods Eighty male C57BL/6J mice were selected and randomly divided into normal Control group(Control),the rest were fed a high-fat diet for 1 month combined with intritoneal injection of low dose STZ,and the model mice with blood glucose≧7.8 mmol/ml were prepared successfully.After modeling,mice were divided into model group(Model),Rub group(Rub,50,100 and 200 mg/kg/d)and positive Control group(Met,200mg/kg/d).Mice in Control group and Model group were intragastrically administrated with 0.1 ml normal saline every day for 10 weeks.The living conditions of mice were observed at fixed time body weight were recorded.After 10 weeks of intervention,serum FBG,OGTT,FINS,TG,T-CHO and LDL-C were determined,and HOMA-IR was calculated.The levels of p-AMPK,GLUT2/4,SOD,MDA,CAT and GSH-Px in liver tissues were detected.It method was used to observe the morphological changes of liver.Results Compared with Control group,the levels of FBG,OGTT,HOMA-IR,TG,T-CHO and LDL-C in Model group were significantly increased(P <0.01,P <0.01,P<0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.05),the contents of p-AMPK and GLUT4 in liver tissue were significantly decreased,the contents of GLUT2 were significantly increased(P <0.01,P <0.01,P <0.05),and the contents of SOD,CAT and GSH-Px in liver tissue were significantly decreased.The content of MDA in liver tissue was significantly increased(P <0.05,P <0.01,P <0.01,P <0.01),liver index was significantly increased(P <0.05),and HE results showed swelling of liver cells and balloon-like changes.Compared with Model group,The serum FBG,OGTT,HOMA-IR,TG,T-CHO and LDL-C in Rub group were significantly decreased(P <0.01,P <0.01,P <0.05,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01,P <0.01),p-AMPK and GLUT4 in liver tissue were significantly increased(P <0.01,P <0.01),GLUT2 expression was significantly decreased(P <0.05),SOD,CAT and GSH-Px expression in liver tissue were significantly increased.MDA expression was significantly decreased(P <0.01,P <0.01,P <0.05,P <0.01),the liver index was significantly decreased(P <0.05),and HE results showed that liver tissue structure was improved to varying degrees.Compared with Met group,FBG,OGTT,HOMA-IR,TG,LDL-C,P-AMPK,GLUT2,MDA and liver index of mice in Rub H group were not statistically significant,serum T-CHO content was significantly decreased(P<0.05),liver GLUT4,SOD,The expression of CAT and GSH-Px were significantly increased(P <0.01,P <0.01,P <0.05,P <0.05).Conclusion Rub has a good hypoglycemic effect on T2 DM mice,which may be due to the activation of AMPK/GLUT2/4 pathway in liver tissue and the improvement of oxidative stress response in liver tissue.
