Objective: Preeclampsia(preeclampsia,PE)is a unique disease in pregnancy and a clinical syndrome of multiple system disorders.It is one of the most serious complications in obstetrics and one of the leading causes of maternal and fetal death.the pathogenesis of PE is still not fully elucidated,and existing studies tend to be associated with placental or trophoblastic ischemia and hypoxia,vascular endothelial injury,oxidative stress,genetic and inflammatory immunomodulatory disorders.Some epidemiological and genetic studies have confirmed that the pathogenesis of preeclampsia is related to susceptible genes and gene polymorphisms,but the underlying mechanism is still elusive due to complex ethnic,geographical and other factors,and so far,there are few clinical studies on preeclampsia gene markers.Therefore,the study of the pathogenesis of preeclampsia from the gene level can provide a reference for the early diagnosis and prevention of preeclampsia.Studies have shown that when the maternal vasoconstriction factor and vasodilator level imbalance,can make the whole body small blood vessels excessive contraction,leading to elevated blood pressure.Angiotensinogen and nitric oxide are common vasoconstrictors and vasodilators.AGT gene can encode and form angiotensinogen,AGT gene polymorphism can lead to the increase of angiotensinogen production in vivo,resulting in the increase of Ang II production in vivo,and then increase the susceptibility to preeclampsia;Nitric oxide synthase3(NOS3)can produce nitric oxide in vascular endothelium and relaxes blood vessels.NOS3 gene polymorphism can lead to the decrease of nitric oxide synthesis / bioavailability,which can lead to endothelial dysfunction and induce preeclampsia.Therefore,this paper will study the correlation between AGT and NOS3 gene polymorphisms and the pathogenesis of preeclampsia.Methods :187 preeclampsia patients were randomly selected from May 2019 to December 2019,A case-control study of 175 normal pregnant women was conducted at random,Using fluorescence probe PCR quantitative method to analyze the correlation between AGT gene polymorphism sites(rs3789679、rs699、rs2004776)and NOS3 gene polymorphism sites(rs3918188、rs1799983),And use R for statistical analysis of data.Results: Through experimental research and data analysis,A genetic balance test was performed on the whole group data and the group data of Hardy-Weinberg case group and control group,AGT rs3789679、rs2004776 accord with Hardy-Weinberg genetic balance(P>0.05),while rs699 matched the Hardy-Weinberg genetic balance in the case group(P>0.05),but not in the overall data and control group(P<0.05);NOS3 rs3918188、rs1799983 all accord with Hardy-Weinberg genetic balance.Analysis of association data for gene polymorphism loci,AGT rs3789679 in the five genetic models of codominant,dominant,recessive,overdominant and log-additive(P>0.05);There were significant differences in AGT rs699 among the four models of codominant,dominant,overdominant and log-additive(P<0.05);There were significant differences in AGT rs2004776 among the four models of codominant,dominant,recessive and log-additive(P<0.05).NOS3 rs3918188、rs1799983 showed no significant difference in the five models(P>0.05).Conclusion: AGT rs2004776 gene polymorphism loci are positively correlated with the pathogenesis of preeclampsia.Although AGT rs699 gene polymorphism loci are correlated with preeclampsia in correlation analysis,the overall data and control group do not conform to the law of Hardy-Weinberg balance.Therefore,whether there is correlation still needs to be further studied.Analysis of the selected NOS3 gene polymorphism sites showed no correlation with preeclampsia.Since this experiment is a small sample experiment and can not exclude external interference factors,the correlation between AGT and NOS3 gene polymorphisms needs further study and practice in large sample and prospective cohort experiments.
