| ObjectiveTo observe the the effect of Yiqi Jianpi TCM compound on gastrointestinal motility and the changes in symptoms of diabetic gastroparesis(DGP)rats,the gastric emptying rate,small intestinal propulsion rate,and pathological changes of the rats in each group were detected,and the Ghrelin,GHSR,CNP,NPR-B,cGMP protein expression.And to expore the the possibile mechanism on gastrointestinal motility in DGP rats.MethodsThe DGP rat model was established by streptozotocin(STZ)combined with high-fat and high-sugar irregular feeding methods.After 6 weeks,the model rats were randomly divided into western medicine group(domperidone group)and traditional Chinese medicine group(Yiqi Jianpi TCM compound Group)and model group.The blank group and the model group were given normal saline 10ml/kg daily by gavage,the traditional Chinese medicine group was given a daily gavage of 5g/kg of Yiqi Jianpi TCM compound,the western medicine group was gavaged with domperidone 3.15×10-3 g/kg every day,and the treatment was lasted for 4 weeks.During the experiments,the rats’ mental state,body shape,mobility,coat color and other general symptoms were scored,and the rats’ fasting blood glucose and body weight changes were recorded.Phenol red gavage method to detect gastric emptying rate and small intestinal propulsion rate,HE staining to observe the pathological changes of gastric antrum tissue,immunohistochemical method to determine the expression of Ghrelin in gastric antrum tissue,WB was used to determine the expression of Ghrelin,GHSR,CNP,NPR-B protein.ELISA was used to detect the expression of cGMP protein in rat gastric antrum tissue.ResultsCompared with the blank group: the weight of rats in the model group decreased(P<0.01),the gastric emptying rate and small intestinal advancement rate were significantly slowed(P<0.01),the gastric antrum tissue Ghrelin and GHSR decreased(P<0.01),the gastric antrum tissue CNP,NPR-B,cGMP expression increased(P<0.01),fasting blood glucose,symptom score increased(P<0.01),HE stained gastric antrum tissue mucosal layer is sparsely arranged,irregular,and glands decreased.Compared with the model group:(1)The fasting blood glucose level of the rats in the Chinese medicine group was significantly reduced,and the difference was statistically significant(P<0.05).The symptom score of the rats in the Chinese medicine group and the western medicine group was reduced(P<0.05).(2)The gastric emptying rate in the Chinese medicine group and the small intestine propulsion rate were significantly accelerated(P<0.05).(3)HE staining showed that the treatment group improved the pathological state of the greater gastric antrum to varying degrees.(4)The expression of Ghrelin and GHSR in the antrum tissue of the Chinese medicine group increased(P<0.05),and the CNP,The expression of NPR-B and cGMP decreased(P<0.05).Compared with the western medicine group:(1)Ghrelin and GHSR expression increased(P<0.05).(2)The traditional Chinese medicine group cGMP decreased,the difference was statistically significant(P<0.05).Conclusions(1)Yiqi Jianpi TCM compound can improve the general state of DGP rats,reduce fasting blood sugar of DGP rats,increase the gastric emptying rate,small intestine advancement rate,and improve gastrointestinal motility in DGP rats.(2)The mechanism of the Yiqi Jianpi TCM compound to improve gastrointestinal motility in DGP rats may be related to the promotion of Gherelin and GHSR expression.(3)Yiqi Jianpi TCM compound improve gastrointestinal motility in DGP rats,which may be related to the regulation of CNP/cGMP signaling pathway and subsequent promotion of gastric smooth muscle contraction. |
