Objective: To observe effect of acupuncturing different acupoints selection based on the location of acupoints related to specific disease for Diabetic Gastroparesis rats, so as to explore whether selecting acupoints according to the location is one of the main factors of influence the effect of acupoints compatibility.Methods: Sixty SPF SD rats of male were randomly divided into blank control group, model group, Zusanli and Zhongwan group, Zusanli and Neiguan group, Zusanli and non acupoint group after 1 week of adaptive feeding, 12 rats in each group. Except the blank control group, DGP model rat was established by injection of 55mg/kg, 2% STZ into peritoneal cavity and raised by common fodder irregularly for 8 weeks, and in the 10 th week to acupuncture treatment and were killed after 4 weeks of treatment. Using ELISA to detect serum ghrelin content; using immunohistochemical detection of gastric fundus tissue and hypothalamus in ghrelin protein content; using real time PCR to detect the gastric fundus tissue, hypothalamic ghrelin and GHS-R m RNA expression.Results: 1.Compared with the blank group, model group of intake, hypothalamus and fundic ghrelin and GHS-R m RNA expression increased significantly, small intestinal propulsion rate, serum ghrelin content, hypothalamus and fundic ghrelin gray value significantly decreased(P < 0.05), indicating that the modeling; 2.Compared with the model group, Zusanli(ST36) and Zhongwan group, Zusanli and Neiguan, Zusanli(ST36) and non the non acupoint group hypothalamus and fundic ghrelin and GHS-R m RNA expression decreased obviously, small intestinal propulsion rate, serum ghrelin content, hypothalamus and gastric ghrelin gray value expression was significantly increased(P < 0.05), indicating that the three treatment groups were effective in the treatment of DGP. 3.Compared with Zusanli and Zhongwan group, Zusanli and Neiguan group and Zusanli and non acupoint group by the increase of the amount of food intake, Zusanli and Neiguan group gastric fundus tissue and hypothalamus of ghrelin and GHS-R m RNA expression were increased(P < 0.05), Zusanli and Neiguan group serum ghrelin levels decreased significantly(P < 0.05), indicating that the Zusanli and Zhongwan group in DGP rat ghrelin regulation is better than that of Zusanli and Neiguan group.Conclusion: Acupuncture could improve the gastrointestinal motility of diabetic gastroparesis rat related serum ghrelin levels and gastric tissue and hypothalamus in ghrelin and GHS-R expression; combined with local acupoints is better than distal acupoints,and acupoint selection is the main factors to influence the effect of acupoints compatibility.
