Objective This study aimed to examine the expression of katanin subunits p60 and p80 in the tissues of human papillary thyroid carcinoma(PTC)(including classical or conventional(CPTC)and follicular(FVPTC)variants)and nodular goiter(NG),so as to explore the possibility that katanin subunits p60 and p80 can be used as potential biomarkers for PTC to distinguish NG.At the same time,we investigated association of katanin p60 and katanin p80 expression with lymph node metastasis in PTC.The correlation of katanin subunits p60 and p80 expression in PTC was also studied.Methods 1.Formalin-fixed paraffin-embedded tissue wax blocks from patients with thyroid diseases who received surgical treatment in the First Affiliated Hospital of Jinzhou Medical University from 2010 to 2020 were selected.Immunohistochemistry was used to examine the expression of katanin subunits p60 and p80 in 97 tissue specimens of benign and malignant thyroid diseases,including 87 PTC(74 of which were CPTC: 39 with lymph node metastasis and 13 FVPTC: 3 with lymph node metastasis)and 10 NG.The relationship between katanin p60 and p80 in PTC lymph node metastasis and the correlation between their expressions were analyzed.2.Cultured thyroid papillary cancer cell KTC-1 in vitro,the katanin p60 protein was overexpressed by transient transfection of exogenous KATNA1 plasmid into KTC-1 cells,and the expression of katanin p80 was detected by Western blot.Results 1.Katanin p60 and p80 were highly expressed in PTC,but not in NG.(p60: P =0.000;p80: P =0.000),Cut-off values for katanin p60 and p80 were 22.43% and 0.83% respectively.2.The expression of katanin p80 in CPTC was significantly higher than in FVPTC(P =0.000).3.The expression of katanin p60 in PTC was significantly associated with lymph node metastasis(P =0.002).4.There was a positive correlation between katanin p60 and katanin p80 expression in PTC(r=0.364,P =0.001).5.Over-expression of katanin p60 significantly enhanced katanin p80 expression in KTC-1 cells(P =0.032).Conclusions 1.The expression of katanin p60 and p80 was found in PTC,but not in NG,which indicated that both of them may be used as potential biomarkers for PTC to distinguish NG.2.In PTC,the high expression of katanin p60 is associated with lymph node metastasis,and the expression of katanin p60 was positively correlated with the expression of katanin p80,providing new potential targets for further research and treatment of PTC. |
