Objective:To predict the target and pathway of Buyang Huanwu Decoction in the treatment of CI by network pharmacology technology,and to further predict its mechanism of action.Methods: In TTD and TCMID databases,seven traditional Chinese medicines including Radix Astragali,Radix Paeoniae Rubra,Rhizoma Chuanxiong,Semen Persicae,Carthami Flos,Uncaria rhynchophylla and Achyranthes bidentata were input respectively,and their active components and targets were obtained.After Swiss Target Prediction database,the target protein was predicted,and finally it was the target of drug action.Get the targets of cerebral infarction diseases from TTD and Dis Ge NET databases,and take the intersection genes of drug diseases,draw the active ingredient-target-disease network diagram of Buyang Huanwu Decoction through the drawing software of Cytoscape3.7.2,build PPI network,obtain key gene targets,network analysis,GO function analysis and KEGG pathway analysis,manually search the literature for pathway annotation,and then analyze the multi-components-multi-targets-of Buyang Huanwu Decoction in treating cerebral infarction.Results: A total of 152 effective active ingredients were obtained from TTD and TCMID databases,and 513 cross genes were obtained from TCMSP,Dis Ge NETSwiss,Target Prediction,String and Unitport.It was predicted that there were 82 targets for treating CI of Paeonia lactiflora,19 targets for Chuanxiong,102 targets for Uncaria rhynchophylla,81 targets for safflower,81 targets for Astragalus membranaceus,55 targets for Achyranthes bidentata and 93 targets for peach kernel.GO enrichment showed that the molecular biological effects mainly occurred in the plasma membrane and exosomes,and inflammatory response and leukocyte migration were the main physiological activities,especially protein binding.KEGG analysis showed that three fluid shear stress and atherosclerosis pathways,tumor necrosis factor signaling pathway and fibrin clot dissolution pathway were enriched.Conclusion: Buyang Huanwu Decoction in the treatment of CI may mainly activate TNF,VEGFA,ICAM1,IL,TLR4,SERPINE1,PI3 K and other factors,specifically involving inflammatory response,leukocyte migration,vascular endothelium,thrombosis,apoptosis,neuroinflammation,immune response,coagulation factors,platelet aggregation,protein binding and a series of ways to promote or inhibit the above reaction and restore hemodynamics And improve the neurological deficit,promote the reperfusion of necrotic area,promote angiogenesis and improve prognosis,play the role of the treatment of CI. |
