Objective:1.This subject intends to use Meta analysis to systematically evaluate the study on the histopathological morphology of Chinese medicine intervention in DMBA-induced breast cancer precancerous lesions in rats.2.Using immunohistochemistry to detect epithelial mesenchymal transition(EMT)-related marker proteins(epithelial marker protein E-cadherin,interstitial marker protein Vimentin)and signaling pathway molecules in human breast benign and malignant histological specimen The expression of JNK provides a new mechanism of action and molecular targets for TCM intervention in breast cancer precancerous lesions.Methods:1.Part 1:Search with Chinese as "Chinese medicine","Breast cancer","Precancerous lesion","Pathology","Animal model","DMBA",and "Rat" as search terms or keywords,and English as PubMed Search,using a computer system to comprehensively search China HowNet,Wanfang,Weipu,Chinese biomedical literature database and PubMed,using Chinese medicine to intervene in DMBA-induced RCT of rat breast cancer precancerous lesion,using Revman5.3 Statistical Analysis.2.Part 2:This study collected breast and thyroid surgery at the Affiliated Hospital of Shandong University of Traditional Chinese Medicine from October 2017 to October 2019 in the Department of Breast Surgery at the Hospital of Shandong University of Traditional Chinese Medicine,Breast ductal epithelial hyperplasia(UDH),breast atypical hyperplasia(ADH),breast ductal carcinoma in situ(DCIS),invasive ductal carcinoma of the breast(IDC)tissue specimens,divided into 4 groups,20 cases each,all female patients.Collect patient’s clinical data in detail,including patient’s age,breast disease history,pathological type,design Excel template and input data,use statistical software SPSS 22.0 software for statistical analysis,perform normality test and homogeneity of variance for each group of experimental data For the test and KW rank sum test,the data is expressed as the median(interquartile range)(Md(IQR)).The correlation between E-cadherin,Vimentin,JNK expression levels and UDH,ADH,DCIS,IDC was analyzed.Results:1.Part 1:The 9 articles included included a total of 327 experimental rats,including 166 in the Chinese medicine group and 161 in the model group.Heterogeneity test results showed that:Chi2=1.77,P=0.99,12=0%,suggesting that there was no heterogeneity between studies,and a fixed effect model was used to combine the effects.The results showed that:RR=0.34,95%CI=[0.21,0.54],P<0.00001,indicating that the difference between the Chinese medicine group and the model group was statistically significant,suggesting that Chinese medicine can block breast cancer in rats modeled by DMBA The role of pre-lesion.2.Part 2:(1)comparing the four groups by KW test,.the P values of E-cadherin score,Vimentin score and JNK score are all<0.05,it is considered that there are at least two groups in E-There are significant differences in cadherin score,Vimentin score and JNK score.(2)Regarding the expression of E-cadherin,there are significant differences between UDH group and ADH group,UDH group and DCIS group,UDH group and IDC group.There are significant differences between ADH group,DCIS group and IDC group.However,the difference between the DCIS group and the IDC group was not significant.In terms of median comparison,the UDH group>ADH group>DCIS group,the UDH group>ADH group>IDC group,there was no statistically significant difference between the DCIS group and the IDC group.(3)Regarding the expression of Vimentin,there are significant differences between UDH group and ADH group,UDH group and DCIS group,UDH group and IDC group.There are significant differences between ADH group,DCIS group and IDC group.However,the difference between the DCIS group and the IDC group was not significant.In terms of median comparison,UDH group<ADH group<DCIS group,UDH group<ADH group<IDC group,there was no significant statistical difference between DCIS group and IDC group.(4)In terms of JNK expression,there were significant differences between UDH group and DCIS group,UDH group and IDC group.There are significant differences between ADH group and DCIS group,ADH group and IDC group.In terms of median comparison,the DCIS group and the IDC group are collectively referred to as the malignant group,the UDH group and the ADH group are collectively referred to as the benign group,and the malignant group<benign group.There was no statistically significant difference between DCIS group and IDC group,UDH group and ADH group.(5)Sort the four pathological tissues in the order of UDH,ADH,DCIS,and IDC(that is,the degree of progression of the disease from light to heavy),count the protein expression levels of E-cadherin and Vimentin in each tissue,and do Spearman correlation Analysis,P=0.000,indicates that E-cadherin and Vimentin have a significant correlation,the correlation coefficient rs=-0.434<0 and the absolute value is greater than 0.4,indicating that there is a significant strong negative correlation between the two.That is,in the process of disease from light to severe,the expression levels of E-cadherin and Vimentin are negatively correlated.Conclusions:1.Part 1:Meta analysis results show that Chinese medicine can effectively intervene to reduce the carcinogenesis of breast cancer precancerous lesions.However,due to the small sample size and poor literature quality,large sample and multi-center RCT studies are still needed to confirm.2.Part 2:From UDH-ADH to malignant tissues,the invasiveness gradually increases,the expression of E-cadherin gradually decreases,and the expression of Vimentin gradually increases.Among them,the expression levels of E-cadherin and Vimentin were negatively correlated,suggesting that the expression of E-cadherin in breast tissue gradually decreased and the expression of Vimentin was gradually increased,which was related to the occurrence and development of cancer tissues and invasive evolution.In the aggressive evolution of breast tissue from benign to malignant,JNK expression gradually decreased. |
