Objective:In this paper,based on the metabonomics,the differen-tial metabolites of Hashimoto’s thyroiditis in TCM syndromes are studied,and the characteristic metabolic pathway is obtained,which provides the basis for the TCM syndrome differentiation and classification of Hashimoto’s thyroiditis,so as to improve the TCM syndrome differentiation and treatment theory system of Hashimoto’s thyroiditis,and enrich the TCM syndrome differentiation methods.Methods:1.From May 2019 to October 2019,54 patients with Hashimoto’s thyroiditis in the Affiliated Hospital of Shandong University of Traditional Chinese medicine were selected for TCM syndrome differentiat-iation and typing,and 10 healthy volunteers were taken as the control.According to the clinical classification,the serum samples were divided into 18 cases of spleen kidney Yang deficiency in group A,18 cases of liver stagnation in group B,18 cases of Yin deficiency and fire hyperactivity in group C and 10 cases of health in Group D;2.The above serum was analyzed by Analysis of ultra high performance liquid chromatography-mass spectrometry and the chromatogram was collec-ted;3.Use progenesis Qi 2.0 analysis software for data preprocessing,and then use SIMCA-P 14.1 the software carries out principal component analys-is,orthogonal partial least square method pattern recognition analysis on the processed data,selects potential metabolites according to the difference weight contribution value(VIP),obtains the differential metabolites according to the difference multiple>1.2,VIP>1.0,P<0.05,and analyzes the metabolic pathway of the differential metabolites using KEGG data platform,The metabolic differences between the serum of Hashimoto’s thyroiditis and the serum of healthy people were studied.Results:1.In the PCA analysis of quality control samples,we can see no outliers,which proves that the experimental system is stable and the data is reliable;2.It can be seen from the total ion diagram that the metabolic differences between different syndromes are obvious3.PCA analysis of the four groups showed that the samples were basically in the 99%confidence interval,and there were differences among the groups.So this experiment can be continued4.There are 8 kinds of different metabolites in PCA principal component analysis between spleen kidney yang deficiency type and healthy group.Among them,5 kinds of metabolites of hippuric acid,citric acid,succinic acid,plant woodcutter alcohol and palmitoyl carnitine are down regulated compared with normal people,and 3 kinds of metabolites of tyrosine,lysophosphatidylcholine and phenylalanine are up regulated.Hashimoto’s thyroiditis of Yang deficiency of spleen and kidney affects human metabolism through 4 metabolic pathways:phospholipid metabolism,phenylalanine metabolism,three carboxylic acid circulation and sphingo-lipid metabolism5.There are 7 groups of differential metabolites between the patients with Hashimoto’s thyroiditis of stagnation of liver Qi type and the healthy people,among which serine,lysophosphatidylcholine,acetylacet-ic acid,pyruvic acid,homocysteine,N-acetylamino 6 metabolites aremetabolites are up-regulated compared with the normal people,and deoxy-d-glucose 1 metabolite is down regulated.HT with stagnation of liver Qi affects human metabolism through four metabolic pathways:phospholipid metabolism,cysteine metabolism,tricarboxylic acid cycle and glucosamine synthesis pathway;6.There are 11 groups of different metabolites between HT of Yin deficiency and and healthy people.Among them,7 kinds of phosphatidylcholine and 8 kinds of oxidized glutathione metabolites are up-regulated compared with normal people,and 2 kinds of alidylcholine and 3 kinds of inosine metabolites are down regulated.The metabolism of Hashimoto’ s thyroiditis of yin deficiency and fire hyperactivity is affected by four metabolic pathways:glycosyl phosphatidy linositol lipid anchor biosynthesis,glycerol phospholipid metabolism,glutathione metabolism and purine metabolism.Conclusion:By using the method of serum metabolomics,it is concluded that there are different metabolites between the three syndrome types and the volunteer’s group;compared with the volunteer’s group,there are 8 different metabolites and 4 different metabolic pathways in spleen kidney yang deficiency type;there are 7 different metabolites and 4 different metabolic pathways in liver qi stagnation type;there are 11 different metabolites and 4 different metabolic pathways in Yin deficiency Huowang type.There are different metabolites among the Three TCM syndromes,and they affect the metabolism of the body through the corresponding metabolic pathway;the same metabolic pathway of spleen kidney yang deficiency,liver depression qi stagnation and yin deficiency fire hyperactivity is phospholipid metabolism;there are two common metabolic pathways of spleen kidney yang deficiency and liver depression Qi Stagnation:phospholipid metabolism and tricarboxylic acid cycle.The main characteristics of differential metabolism are lipid metabolism,amino acid metabolism and tricarboxylic acid cycle.From the perspective of metabonomics,this study confirmed that there were differences in metabolites and metabolic pathways between different TCM Syndromes of Hashimoto’s thyroiditis,which provided a basis for the objectification of TCM syndrome differentiation of this disease.
