| As the four properties of Traditional Chinese Medicine(TCM)is the core of the Chinese materia medica property theory,modern research methods applied in the traditional theory research and interpretation of the scientific connotation has important significance to promote the modernization of TCM theory.In this paper author chooses typical cold property of medicine(radix scutellariae,rhizoma coptidis,cortex phellodendri)and typical hot property of medicine(aconite,dried ginger,Zanthoxylum bungeanum Maxim)as the research object in order to find representative biomarkers of four properties by constructing the methodology framework based on metabonomics research,which will prove the hypothesis(Four properties are biological effects of TCM that influence the body’s energy metabolism and substance metabolism through various ways characterized with therapeutic relevant or not,as the same time influence curative effect or associate with side effects)and expounds the scientific connotation of the four properties.Based on the concept,this paper has carried out the research in following aspects:1)The literature review.Search through a large number of literature at home and abroad on the four properties and metabonomics research and the relationship between the two to carry out literature foundation of this paper.2)Research on the effect of typical cold and heat drugs which act to normal rats’substance metabolism and energy metabolism based on metabonomics to find out the biomakers and pathways that have a connection with four properties;3)Research on the effect of typical cold and heat drugs which act to cold syndromes rats’ substance metabolism and energy metabolism based on metabonomics to find out the biomakers and pathways that have a connection with four properties;4)Research on the effect of typical cold and heat drugs which act to heat syndromes rats’ substance metabolism and energy metabolism based on metabonomics to find out the biomakers and pathways that have a connection with four properties.This research found some biomarkers such as tetrahydropteridine,phosphorylcholine and so on which may be the point of action of typical cold traditional Chinese medicines and its.the related pathways such as glycosphingolipid metabolism,metabolism of vitamin and so on.Also it found some specific point such as 13S-hydroxyoctadecadienoic acid and ascorbic acid and its related metabolic pathways such as linoleic acid metabolism,PPAR signaling pathways and other metabolic pathways which could be biomarkers of action of typical hot traditional Chinese medicines.Then it discoveries 20 biomarkers that both testing medicine aimed at such as isocitric acid,stachyose,indoleacrylic acid,D-Proline and so on,which are involved in metabolic pathways of energy substances such as glucose metabolism,amino acid metabolism and sphingolipid metabolism,and metabolic pathways of key enzymes such as Pantothenate and CoA biosynthesis,which also involved in important energy metabolic pathways such as the Krebs cycle.The result shows that the typical cold,hot Chinese medicines have different influences and effect of different trends on biomarkers and metabolic pathways to show the influence on body’s energy metabolism and material metabolism.Comprehensive results show that this study preliminary demonstrated the hypothesis of the four properties that put forward in early stage of the research and it improved the method framework of research on four properties,which has an important theoretical significance and application value. |
