Based On The “Brain-Gut-Bacteria” Axis To Explore The Effect Of Liqi Tongbian Prescription On Brain Gut Peptide And Intestinal Flora Of Patients With Qi Stagnation Syndrome Of Slow Transit Constipation

Objective:The correlation between slow transit constipation and “brain-gut-bacteria”axis was verified by clinical observation,with a view to exploring the pathogenesis of slow transit constipation,and the curative effect of Liqi Tongbian prescription on slow transit constipation(qi stagnation syndrome)and regulating the “brain-gut-bacteria” axis,which would provide new ideas for improving the clinical efficacy.Methods:In this study,a total of 60 patients with slow transit constipation(qi stagnation syndrome)were randomized into TCM treatment group and western medicine control group according to random digit table,and another 20 healthy people were set as normal control group.TCM treatment group:the treatment of Liqi Tongbian prescription(1 dose/d,two times a day).Western medicine control group:the medication was administered mosapride tablets(5mg,three times a day)and wheat cellulose granules(3.5g,three times a day).Consecutively for 4 weeks.Recording qi stagnation syndrome patients of the symptoms score of constipation,and constipation severity score,SCBM of per week and the 72 h colon excretion rate before and after treatment.5-THR3、5-THR4、MTL、NO、SP and intestinal flora were tested before-after treatment of patients and normal control group.And follow up the patients with definite efficacy after 1 month after treatment,calculate the recurrence and adverse reaction.Results:The clinical curative total effect of the TCM treatment group was better than that of the western medicine control group(P<0.01),the TCM treatment group was better than the western medicine control group(P<0.05)in the patients with “course of disease <5 years” and “course of disease >10years ”(P<0.05),the TCM treatment group was better than the western medicine control group(P<0.05)in the patients with “mild” and “moderate” patients(P<0.05).The qi stagnation syndrome score of constipation and total score of CSS in the TCM treatment group after treatment were lower than those in the western medicine control group(P<0.05).The number of SCBM in the TCM treatment group was more than that in the western medicine control group in the two points in time after treatment(P<0.05).The 72 h colon excretion rate in the TCM treatment group was more than that in the western medicine control group after treatment(P<0.05).Levels of 5-THR3,5-THR4,SP and MTL were higher than those in western medicine control group,whereas level of NO was lower than that in western medicine control group(P<0.05).The relative abundance of intestinal flora in the TCM treatment group was significantly lower than that before treatment(P<0.05),and the diversity was lower than that in western medicine control group(P<0.05).The recovery rate in the TCM treatment group was 17.86%,which was lower than 46.15% in western medicine control group(P<0.05).There were no significant adverse events in the two groups during the treatment and 1 month follow-up,which was safe and reliable.Conclusions:1.Liqi Tongbian prescription treating slow transit constipation(qi stagnation syndrome)can significantly improve the symptoms of constipation,increase the number of SCBM,promote colon movement,regulate brain intestinal peptide and intestinal flora,the clinical effect is better than western medicine control group,and has the characteristics of low recurrence rate,safe and reliable,it is worth popularizing in clinic.2.The pathogenesis of slow transit constipation has a certain correlation with the“brain-gut-bacteria” axis,Liqi Tongbian prescription has a certain effect on patients with slow transit constipation(qi stagnation syndrome),and can regulate the axis.
